摘要:目的 :研究附子水煎剂 (DCMR)对主动脉环的舒张作用及机制。方法 :采用家兔离体主动脉平滑肌标本 ,以去甲肾上腺素 (NA)预收缩主动脉后给予不同剂量的附子观察其张力变化 ,并探讨去除血管内皮、给予NO合酶抑制剂左旋硝基精氨酸 (L NNA)、甲烯蓝 (MB)、环氧酶抑制剂吲哚美辛和 β肾上腺素能受体阻断剂普萘洛尔对血管张力变化的影响。 结果 :附子对血管环静息张力无明显影响 ,但不同剂量的附子可使 10 6mol/LNA预收缩血管产生明显舒张 (r =0 .87,P <0 .0 5 )。去除血管内皮、10 4mol/LL NNA或 10 5mol/LMB可减弱附子的舒张作用 ,但吲哚美辛和普萘洛尔无影响。另外 ,4mg/mL附子水煎剂对无内皮细胞血管环NA(PD2 为 7.0 7± 0 .90和 6.93± 0 .87,P >0 .0 5 )和KCl(PD2 为 1.40± 0 .3 6和 1.2 5± 0 .2 1,P >0 .0 5 )的量效收缩反应无明显影响。结论 :附子水煎剂对主动脉的舒张作用是内皮依赖性的 ,且与内皮释放的NO有关 ;而与平滑肌细胞膜上的受体依赖性Ca2 + 通道和电压依赖性Ca2 +
Objective: To study vasodilation effect of a water decoction of the Common Monkshood Prepared Daughter Root(DCMR) on rabbit thoracic aortic rings and its mechanism. Method: In noradrenaline(NA) precontracted isolated aortic rings in rabbits, the response to different concentration of DCMR was investigated; the effects of removal of endothelium, coincubated with nitric oxide synthase inhibitor L NNA, methylene blue(MB), prostaglandin synthase inhibitor and blockage of the adrenergic β receptor on DCMR induced relaxation were also assessed. Result: DCMR did not change the resting tension of rabbit aortic rings but DCMR with different concentration could cause an obvious relaxation in 10 6 mol/L NA precontracted aortic rings(r=0.87,p<0.05).The relaxant effect of DCMR was significantly reduced by removal of endothelium 10 4 mol/L L NNA and 10 5mol/L MB, but not by prostaglandin synthase inhibitor and blockage of the adrenergic β receptor (Prop). In addition DCMR(1mg/mL)did not inhibit NA and KCl cumulative concentration response curves of aortic rings without endothelium, and did not change the PD 2 values for NA from(7.07±0.90)in control group to (6.93±0.87,P>0.05)and for KCl from(1.40±0.36) in control group (1.25±0.21) (P>0.05) respectively. Conclusion: DCMR can relax isolated rabbit aorta, which may be endothelium dependent and involve in nitric oxide (NO), but is not related to blockage of receptor operated and voltage dependent calcium channels.
