摘要:目的:比较高乌头生品和炮制品的镇痛作用及其中总生物碱与高乌甲素的含量。方法:采用小鼠热板法和扭体法对高乌头炮制前后的镇痛作用进行研究;分别采用酸性染料比色法和高效液相色谱法测定样品中总生物碱和高乌甲素的含量。结果:在扭体法实验中,高乌头炮制前后的镇痛率分别为16.84%和50.52%,炮制品明显高于生品,但无统计学意义;在热板法实验中,给药后30 min、45 min、60 min,高乌头生品及炮制品的痛阈提高百分率分别为37.78%、74.34%、109.05%及76.04%、111.59%、111.13%,与正常对照组比较差异显著(P<0.05),但炮制品与生品比较差异不显著。高乌头炮制前后总生物碱和高乌甲素的质量分数分别为2.06%、7.75%和1.170%、0.596%。结论:高乌头炮制后镇痛作用增强,表明以甘草汁炮制能够增强高乌头的镇痛作用,且是通过增加总生物碱的含量,而不是增加高乌甲素的含量而达到增效作用。
Objective: To compare the analgesic effect and the contents of total alkaloids and Lappaconitine in the unproe.essed Aconitum sinomontanum Nakai and processed product, and to provide a basis of for discussion of processing mechanism of A conitum sinomontanum Nakai. Methods: The analgesic effect test of Aconitum sinomontanum Nakai was carried out by hot-plate test and writhing test in mice, and the determination of alkaliods and Lappaconitine in the two samples was performed by acidic dye colorimetry and high performance liquid chromatography (HPLC), respectively. Results: In mice, the analgesia percentage treated with the unprocessed sample and the processed one was 16.84% and 50.52%, respectively, for the writhing test. Although the analgesia percentage of the processed was obviously higher than that of the unprocessed, there was not statistically significance. The improved analgesic threshold of the unprocessed sample and the processed one was 37.78%, 74.34%, 109,05%, 76.04%, 111.59% and 111.13%, respectively, for the hot-plate test at 30, 45, 60 min after drug administration. The pain threshold of mice treated with the processed sample and the unprocessed one was significantly increased (P〈0.05) compared with the normal control group. Although improved percentage of pain threshold of the processed product was obviously higher than that of the unprocessed, there was not statistically significance. The contents of the general alkaloid and Lappaconitine in the unprocessed product and the processed one was 2.06%, 7.75% and 1.170%, 0.596%, respectively. Conclusion: The analgesic effect of processed Aconitum sinomontanum Nakai is higher than that of the original rude drug. It showed that the analgesic effect of Aconitum sinomontanum Nakai can be improved by processing with licorice juice, and the increasing analgesic effect of the processed product is achieved by enhancing the content of general alkaloids, but not increasing the content of Lappaconitine.
