摘要:目的探讨黄芪多糖联合二甲双胍治疗衰老糖尿病的分子机制。方法高糖高脂饲料联合链脲佐菌素诱导自然衰老小鼠建立衰老糖尿病模型,将实验小鼠分为衰老对照组、衰老糖尿病模型组、二甲双胍治疗组、黄芪多糖联合二甲双胍治疗组。检测各组小鼠血清胰岛素变化,胰腺组织胰岛素分泌情况及胰腺组织的形态学变化;利用生物信息学工具分析衰老对照组、衰老糖尿病模型组、黄芪多糖联合二甲双胍治疗组小鼠的肝脏组织差异mRNA功能。结果黄芪多糖联合二甲双胍治疗相比二甲双胍单药组可显著升高衰老糖尿病小鼠的血清胰岛素含量(P<0.05),改善胰腺腺泡细胞水肿和促进胰岛形态的恢复。芯片结果显示,差异mRNA,Tob2、Tmem100及Usp15等显著差异表达且对它们的靶miRNA具有较强调控作用。经过功能注释,发现差异mRNA在BMP信号通路、cGMP-PKG信号通路及GnRH信号通路中显著富集。结论黄芪多糖联合二甲双胍可能通过调控BMP信号通路、cGMP-PKG信号通路,Tob2,Usp15及Tmem100基因改善肝脏组织损伤,对缓解衰老糖尿病所致损伤具有研究潜能。
Objective To explore the molecular mechanism of astragalus polysaccharide combined with metformin in the treatment of aging type 2 diabetes. Methods Establish the senescence diabetes model of mice induced by high-sugar and high-fat diet combined with streptozotocin, and divide the experimental mice into senescence control group, senescence diabetes model group, metformin treatment group, and astragalus polysaccharide combined with metformin treatment group.The changes of serum insulin, insulin secretion in pancreatic tissue and morphological changes of pancreatic tissue in each group were detected.Bioinformatics tools were used to analyze the different mRNA functions of liver tissues in aging control group, aging diabetes model group, astragalus polysaccharide combined with metformin treatment group. Results Compared with the metformin group, aps combined with metformin treatment significantly increased the serum insulin content in aging diabetic mice(P<0.05), improved the acellular edema of pancreas and promoted the recovery of pancreatic islet morphology.Chip results showed that the differentially expressed mRNA, Tob2, Tmem100 and Usp15 had a strong regulatory effect on their target miRNA.Through functional annotation, it was found that differential mrnas were significantly enriched in the BMP signaling pathway, cgmp-pkg signaling pathway and GnRH signaling pathway.Conclusion Astragalus polysaccharide combined with metformin mainly improves liver tissue injury by regulating BMP signaling pathway, cGMP-PKG signaling pathway and Tob2, Usp15 and Tmem100 have research potential to alleviate the injury caused by aging diabetes.
