摘要:目的观察钩藤碱对自发性高血压大鼠(SHR)血压和肠道菌群的影响。方法将50只8周龄SHR随机分为模型组(Model组,n=10)、卡托普利组(CAP组,12.5 mg·kg^(-1)·d^(-1)卡托普利灌胃,n=10)、钩藤碱高剂量组(Rhy-H组,10 mg·kg^(-1)·d^(-1)钩藤碱灌胃,n=10)、钩藤碱中剂量组(Rhy-M组,5 mg·kg^(-1)·d^(-1)钩藤碱灌胃,n=10)、钩藤碱低剂量组(Rhy-L组,2.5 mg·kg^(-1)·d^(-1)钩藤碱灌胃,n=10),另设Wistar大鼠作为正常对照组(NC组,n=10)。正常对照组和模型组每日给予生理盐水灌胃,干预6周后测定各组大鼠血压,实验结束前取大鼠粪便,进行16S rDNA V3-4去扩增子信息采集与分析。结果钩藤碱对自发性高血压大鼠具有明显的降压作用。正常对照组、模型组、卡托普利组和钩藤碱高、中、低剂量组的Alpha多样性指数的Chao1指数分别为634.09±26.62,623.09±14.41,643.08±26.40,640.38±22.90,673.95±26.06和610.41±31.16;Ace指数分别为629.18±28.74,612.72±22.04,642.54±22.75,635.61±30.94,662.87±10.74和596.87±27.02;Shannon指数分别为4.36±0.46,4.24±0.19,3.89±0.17,4.02±0.26,4.25±0.21和3.80±0.27;Simpson指数分别为0.06±0.05,0.05±0.02,0.08±0.01,0.07±0.02,0.05±0.01和0.09±0.03。在门水平,与模型组比较,卡托普利、钩藤碱高剂量组以及钩藤碱低剂量组Firmicutes丰度降低,Bacteroidetes丰度增加,差异有统计学意义(P<0.05)。在属水平,与模型组比较,钩藤碱高剂量组Ruminococcus丰度明显减少;钩藤碱低剂量组Oscillospira、Ruminococcus丰度明显减少,Prevotella丰度显著升高,差异均有统计学意义(P<0.05,P<0.01)。结论钩藤碱具有明确的降压作用,并能一定程度上改善肠道菌群紊乱,优化肠道菌群组成。
Objective The study investigated the influence of rhynchophylline(Rhy)on blood pressure and gut microbiota in spontaneously hypertensive rats(SHR)by 16 S rDNA sequencing.Methods 8-week-old male SHR(n=50)were randomly divided into 5 groups:model group,captopril group(CAP group,12.5 mg·kg^(-1)·d^(-1)CAP by gavage),Rhy-H group(10 mg·kg^(-1)·d^(-1)Rhy,n=10),Rhy-M group(5 mg·kg^(-1)·d^(-1)Rhy,n=10),Rhy-L group(2.5 mg·kg^(-1)·d^(-1)Rhy,n=10).Wistar rats were used as the normal control group(NC group,n=10).The rats in NC group and model group were given the same amount of 0.9%NaCl once a day by gavage.After 6 weeks,the blood pressure of all rats were measured.The molecular ecology of the fecal gut microbiota was analyzed by 16S r DNA sequencing.Results Rhy decreased blood pressure steadily.The Chao1 index of Alpha diversity index in normal control group,model group,CAP group,Rhy-H,Rhy-M and Rhy-L groups were 634.09±26.62,623.09±14.41,643.08±26.40,640.38±22.90,673.95±26.06,610.41±31.16,respectively;Ace index in the 6groups were 629.18±28.74,612.72±22.04642.54±22.75,635.61±30.94,662.87±10.74,596.87±27.02,respectively;Shannon index in the 6 groups were 4.36±0.46,4.24±0.19,3.89±0.17,4.02±0.26,4.25±0.21,3.80±0.27,respectively;Simpson index in the 6 groups were 0.06±0.05,0.05±0.02,0.08±0.01,0.07±0.02,0.05±0.01,0.09±0.03,respectively.At the Phylum level,compared with model group,the abundance of Firmicutes decreased significantly as well as the abundance of Bacteroidetes increased significantly in CAP group and Rhy-H group,Rhy-L group,the difference of the factors were significant(P<0.05).At the Genus level,compared with model group,the abundance of Ruminococcus decreased significantly in Rhy-H group,the abundance of Oscillospira,Ruminococcus decreased significantly as well as the abundance of Prevotella increased significantly in Rhy-L group,the difference of the factors were significant(P<0.05,P<0.01).Conclusion The intervention with Rhy has a good antihypertensionsive effect and optimizes the gut microbiota structure in rats.
