摘要:SIP合胞体是维持正常胃肠动力的关键,功能性消化不良(FD)胃肠动力障碍与SIP合胞体中ICC-ANO1/PDGFRα+细胞-SK3途径的失衡有关。肝失疏泄、脾胃升降失常是FD发病的基本病机,疏肝健脾法是治疗FD的重要治法。前期研究发现疏肝健脾法改善FD胃肠动力与其调节胃肠肌电活动和脑肠肽释放有关,但是否通过调控SIP合胞体以干预FD目前尚不明确。因此我们提出:“疏肝健脾法通过ICC-ANO1/PDGFRα+细胞-SK3途径调控SIP合胞体促进FD胃肠动力”的科学假说。基于ICC-ANO1/PDGFRα+细胞-SK3途径,采用细胞学、分子生物学和全细胞膜片钳等方法,探讨疏肝健脾法中药含药血清对ICC/PDGFRα+细胞的影响;建立FD胃肠动力障碍大鼠模型,明确疏肝健脾法的干预作用,采用分子生物学和电生理技术验证疏肝健脾法调控SIP合胞体改善FD胃肠动力障碍及其作用机制。
Functional dyspepsia(FD)is associated with the imbalance of ICC-ANO1/PDGFRα+cell-SK3 pathway in SIP syncytium which is the key to maintain normal gastrointestinal motility.Disorder of normal physiological function of liver and spleen is the basic pathogenesis of FD.Soothing liver and invigorating spleen method is an essential treatment concept for FD.Additionally,the previous study of the research group has found that the improvement of gastrointestinal motility in FD by soothing liver and invigorating spleen method is related to the regulation of gastrointestinal myoelectrical activity and the release of brain-intestinal peptides.However,it is currently unclear whether the intervention in FD can be achieved by regulating SIP syncytial.Therefore,this research has proposed the scientific hypothesis that soothing liver and invigorating spleen method promotes the gastrointestinal motility of FD by regulating SIP syncytium through the ICC-ANO1/PDGFRα+cell-SK3 pathway.Based on the ICC-ANO1/PDGFRα+cell-SK3 pathway and used cytology,molecular biology and whole-cell patch-clamp methods,the present project will investigate the effect of serum containing traditional Chinese medicine on ICC/PDGFRα+cells by soothing liver and invigorating spleen method.A rat model of FD gastrointestinal motility disorder was established to clarify the intervention effect of soothing liver and invigorating spleen method.The molecular biology and electrophysiological techniques were used to verify the improvement of FD gastrointestinal motility disorder by regulating SIP syncytium and the related mechanisms.
