摘要:目的观察胰升糖素样肽-1(GLP-1)对2型糖尿病大鼠胰岛细胞bcl-2和bax蛋白表达的影响及与剂量的关系。方法采用高脂高糖饲料喂养结合小剂量链脲佐菌素注射法成功建立2型糖尿病大鼠模型,用高、中、低3个剂量GLP-1对2型糖尿病大鼠进行为期4周的干预,比较干预各组与未干预组糖尿病大鼠胰岛细胞bcl-2和bax蛋白表达的变化。结果对比糖尿病组,GLP-1干预各组大鼠胰岛细胞bax蛋白表达率均明显降低,与GLP-1剂量呈负相关;bcl-2/bax比值均明显升高,与GLP-1剂量呈正相关。结论GLP-1能明显降低2型糖尿病大鼠胰岛细胞bax蛋白的表达率,增加bcl-2/bax比值。提示GLP-1对2型糖尿病大鼠的胰岛细胞有明显抗凋亡作用,并与GLP-1剂量有关。
Objective To investigate the effects of glucagon-like peptide-1(GLP-1) intervention on the expression of bcl-2 and bax protein in islet cell of type 2 diabetes rats.Methods Type 2diabetes rats models was successfully established by feeding high-fat,high-sugar diet and injecting low-dose streptozotocin.Type 2 diabetes rats had been interfered by high dose,middle dose and low dose GLP-1 for 4 weeks.The expressions of bcl-2 and bax protein in islet cell of type2 diabetes rats were compared between GLP-1 intervention groups and no intervention group.Results Intervention groups was significantly lower.The ratio bcl-2/bax of GLP-1 intervention groups was significantly higher,and it was related to the GLP-1 doses.Conclusion GLP-1 can significantly reduce bax protein expression of the type 2 diabetes rats' islet cells,increase the ratio bcl-2/bax and relate to the GLP-1 dose.GLP-1 has significant anti-apoptosis effect on islet cell of type 2 diabetes rats,and it is dose-related.
