摘要:目的:通过观察当归挥发油对自发性高血压大鼠(SHR)ACE2/[Ang(1-7)]/Mas受体轴表达的影响,探讨当归挥发油降压的作用机制。方法:将40只8周龄雄性SHR随机分为模型对照组、低剂量当归治疗组、高剂量当归治疗组、卡托普利治疗组,每组8只;并用同周龄的Wistar大鼠作为空白对照组。各组大鼠分别灌胃4周后,ELISA法检测血清中肾素、Ang(1-7)水平,QT-PCR法检测Mas mRNA表达量,Western blotting检测Mas蛋白的表达水平。结果:与模型对照组相比,低剂量当归治疗组、高剂量当归治疗组血清肾素水平均有所降低,但差异无统计学意义(P>0.05),低剂量当归治疗组、高剂量当归治疗组Ang(1-7)表达水平均较模型对照组升高,但差异无统计学意义(P>0.05),低剂量当归治疗组、高剂量当归治疗组Mas受体mRNA及蛋白表达水平均较模型对照组明显升高,差异有统计学意义(P<0.05)。结论:当归挥发油对SHR血压有调节作用,可能通过ACE2/[Ang(1-7)]/Mas受体轴拮抗ACE系统发挥降压作用。
Objective:To study the influence of Angelica sinensis(oliv.)Diels on ACE2/Ang1-7/Mas receptor in spontaneously hypertensive rats(SHR).Method:All the rats were divided into Angelica group,model control group,captopril group,weekly-old Wistar rats as normal control group,then measure the systolic blood pressure of the tail in different groups before and after treatment.After 4weeks,cardiac tissue of the rats will be taken out.The plasma rennin and Ang(1-7)were evaluated by ELISA.The mRNA levels of Mas was detected by RTPCR,and the protein expressions of Mas was tested by Western blot.Result:Angelica can reduce blood pressure levels in SHR.The level of peripheral plasmarennin was lower and the level of Ang(1-7)was higher than model group,but no significant difference(P〉0.05).The mRNA and protein expressions of Mas was significant increase in Angelica group(P〈0.05).Conclusion:the essential oil of Angelica sinensis can reduce the blood pressure through decrease the level of peripheral plasma rennin and up regulation the expression of ACE2/Ang1-7/Mas receptor axis.The effects of Angelica related to this axis.
