摘要：目的探讨电针"三阴穴"对慢性非细菌性前列腺炎大鼠模型的免疫调节机制。方法将48只雄性SD大鼠随机分为空白组、模型组、药物组、电针组,皮内注射同种异体雄性SD大鼠前列腺蛋白提纯液,辅以双重免疫佐剂复制自身免疫性慢性非细菌性前列腺炎大鼠模型。造模后电针组、药物组分别给予电针"三阴穴"、舍尼通灌胃治疗15 d。观察各组动物前列腺组织湿重及系数、形态结构;放免法检测前列腺组织中抗体Ig G、局部组织细胞因子白介素(IL)-2、IL-8的变化。结果造模后模型组前列腺重量及系数与空白组相比显著增高(P<0.05),病理观察显示,模型组动物前列腺组织形态及结构明显受损。与空白组比较,模型组大鼠前列腺组织匀浆中Ig G含量明显增高(P<0.05),IL-2、IL-8高表达(P<0.05,P<0.01)。治疗后,电针组动物腺体湿重及系数均明显下降(P<0.05,P<0.01),病理变化明显减轻;局部前列腺组织Ig G含量明显降低(P<0.05),IL-2、IL-8表达显著减轻(P<0.05,P<0.01)。结论电针"三阴穴"能够减少局部抗体Ig G的分泌,调节局部细胞因子IL-2、IL-8的表达,减轻组织局部免疫亢进,抑制前列腺组织形态结构的损伤,从而起到治疗慢性非细菌性前列腺炎的作用。
Objective To study the immunologic mechanism of electroacupuncture at Sanyin points on chronic abacterial prostatitis rat. Methods 48 male rats were divided randomly into normal,model,drug and electroacupuncture groups,12 rats in each group. The animal models were made by SC purified prostate protein twice with immunologic adjuvant,and the rats in electroacupuncture and drug groups were treated by electroacupuncture at Sanyin points and fed Cernilton for 15 d. The changes of prostate weight,coefficient,pathology and contents of Ig G,expressions of IL-2 and IL-8 in prostate homogenate were observed. Results Compared with those of normal group,the prostate weight and coefficient were significantly increased in model group( P < 0. 05),the content of Ig G was significantly increased( P < 0. 05),the levels of IL-2,IL-8 were highly expressed( P < 0. 05,P < 0. 01),prostate tissue’s morphology and structure were significantly impaired in model group; compared with those of model group,the prostate weight and coefficient were significantly decreased in electroacupuncture group after treatment( P < 0. 05 or P < 0. 01),pathological changes were reduced significantly,the content of Ig G was decreased significantly( P <0. 05),the expressions of IL-2,IL-8 were significantly decreased( P < 0. 05 or P < 0. 01),and the pathological manifestation of chronic inflammation was significantly improved. Conclusions Electroacupuncture at Sanyin points could effectively improve the immune function of chronic abacterial prostatitis and promote the recovery of inflammation.
