摘要:目的研究黑逍遥散对氢溴酸东莨菪碱所致老年性痴呆(Alzheimer’s Disease,AD)模型小鼠学习记忆及抗氧化能力的影响。方法将60只SPF级小鼠随机分为5组,即空白对照组、模型对照组、黑逍遥散组、逍遥散组及精简方组,每组12只。空白对照组、模型对照组给予生理盐水,黑逍遥散组、逍遥散组及精简方组分别给予相应生药水煎液,连续灌胃45d,其中给药第41~45d,除空白对照组外,其余各组小鼠均腹腔注射氢溴酸东莨菪碱2.0mg/(kg·d)进行造模,造模半小时后进行水迷宫学习能力训练,连续训练5d;造模完成后间隔1d,即第47d进行水迷宫记忆能力测试。学习记忆能力测试完毕后,处死小鼠,取其脑组织,测定小鼠脑组织匀浆中超氧化物歧化酶(SOD)活性及丙二醛(MDA)含量。结果黑逍遥散、逍遥散及精简方均可显著提高小鼠学习记忆能力及脑组织匀浆中SOD活性,降低潜伏时间及MDA含量,但以黑逍遥散作用最为明显。结论黑逍遥散对氢溴酸东莨菪碱引起的小鼠学习记忆能力障碍有明显的改善作用,其作用可能与抗自由基氧化有关。
Objective To research the effects of Heixiaoyao San on study, memory and antioxidation ability of mice with AD induced by scopolamine hydrobromide. Methods 60 mice were randomly divided into blank control group, model control group, Heixiaoyao San group, Xiaoyao San group and Jingjian Fang group, every group had 12 mice. The mice of blank control group and model control group were given normal saline, the mice of other groups were given relevant herbs decoction. All mice had been administrated for 45 days. In the forty first to forty fifth day, except blank control group, the mice of other groups were injected scopolamine hydrobromide of 2.0 mg/( kg · d) in abdominal cavity to make the model of AD. Half hours after making the model, the learning capability of all mice had been trained in water maze for 5 days. On the second day after making the model, tested the memory capability of all mice in water maze. And killed all mice, determined the activity of SOD and the content of MDA in brain tissue of mice. Results Heixiaoyao San, Xiaoyao San and Jingjian Fang could significantly increase the learning and memory capability and the activity of SOD in brain tissue of mice, and decrease latent time and the content of MDA, but the effect of Heixiaoyao San was strongest. Conclusion Heixiaoyao San has obvious protecting effect to the disturbance of learning and memory ca- pability of mice induced by scopolamine hydrobromide,the effect could relate to antioxidation of free radical.
