摘要:为选出培养半夏试管珠芽的适宜Fe^(2+)浓度,该研究通过梯度改变1/2MS基本培养基中Fe^(2+)浓度,依据试管珠芽的形成时间、诱导率,试管苗地上、地下形态指标,试管珠芽中淀粉粒数量与分布,叶片叶绿素含量及试管珠芽有效成分含量等指标,得出试验结果为9.2×10^(-5)mol·L^(-1)浓度下各项指标最佳。此外,还发现Fe缺素下无法形成完整试管珠芽,说明Fe是半夏试管珠芽形成过程中必不可少的微量元素,但同时浓度不宜过高,因高浓度Fe^(2+)下,形成的试管珠芽极小且后续无法正常萌发成苗。
To obtain an optimal Fe^(2+)concentration for culturing test tube bulbils of Pinellia ternata,the concentrations of Fe^(2+)in 1/2MS basic medium were changed in gradient;the formation time,induction rate and the above-ground and underground morphological indices of test-tube plantlets were investigated.The number and distribution of starch granules and the content of chlorophyll in leaves and the content of effective components in test tube bulbils were assayed.The results showed that the optimal Fe^(2+)concentration in 1/2MS basic medium for culturing test tube bulbils was 9.2×10^(-5) mol·L^(-1).In addition,the complete bulbils could not be formed under Fe deficiency.This indicated that Fe was an essential trace element in growth of P.ternata.However,the bulbils formed under high concentration of Fe^(2+)were very small and failed to germinate normally.
