摘要:目的:观察敦煌医学古方大补脾汤对放射性肺损伤大鼠的影响,探讨可能的作用机制。方法:40只SPF级Wistar雄性大鼠随机分为正常对照组,模型组,大补脾汤低、中、高剂量组,每组8只。大补脾汤各组预先灌胃不同剂量大补脾汤(6.07、12.14、24.28g/kg),正常对照组和模型组分别灌胃等量0.9%氯化钠溶液。2周后模型组、大补脾汤各组大鼠右肺予8Gy X射线单次照射,正常对照组不予照射。照射后7d处死大鼠,HE染色观察大鼠右肺组织病理改变,检测外周血细胞含量变化,ELISA检测大鼠血清中TGF-β1、IL-1β、IL-6、TNF-α、IL-8浓度,计算胸腺、脾脏指数。结果:与正常对照组比较,模型组外周血白细胞、红细胞、血小板、血红蛋白等含量显著下降(P<0.05),血清中TGF-β1、IL-1β、IL-6、TNF-α、IL-8浓度显著升高(P<0.05),胸腺指数和脾脏指数均显著下降(P<0.05),HE染色显示大鼠右肺肺泡壁增厚,肺泡腔内有渗出物及炎性细胞浸润等病理改变。与模型组比较,大补脾汤中、高剂量组外周血白细胞、红细胞、血小板、血红蛋白含量等显著上升(P<0.05),大补脾汤各组血清中TGF-β1、IL-1β、IL-6、TNF-α、IL-8浓度显著下降(P<0.05),胸腺指数和脾脏指数显著升高(P<0.05),大鼠的肺组织病理损伤有一定改善。结论:大补脾汤对放射性肺损伤大鼠有一定的防护作用,其机制可能与降低TGF-β1、IL-1β、IL-6、TNF-α、IL-8的表达有关。
Objective:To discuss about the effects of Dabupi Decoction of ancient prescription of Dunhuang medicine on the model rats of radiation-induced pulmonary injury and explore the possible mechanism.Methods:Forty SPF Wistar male rats were randomly divided into 5 groups:the normal control group,the model group,low dose,middle dose and high dose of Dabupi Decoction group.Every Dabupi Decoction groups were given Dabupi Decoction by gavage(6.07,12.14,and 24.28 g/kg).And the normal group and the model group were given the equal saline solution for two weeks in advance.Later the right lungs of the rats of the model group and every Dabupi Decoction groups were radiated once by 8 Gy X-ray,while the normal control group was not radiated.Seven days after radiation all groups of rats were executed.The changes of right lung tissue pathology morphology of the rats were observed by hematoxylin-eosin staining.Thymus index and spleen index,blood routine and the levels of TGF-β1,IL-1β,IL-6,TNF-αand IL-8 in serum of the rats were examined.Results:Compared with the normal control group,the amount of leukocyte,erythrocyte,blood platelet and hemoglobin etc.and thymus index and spleen index of the model group distinctly declined and the level of TGF-β1,IL-1β,IL-6,TNF-αand IL-8 in the serum distinctly increased(P<0.05);HE staining showed that the alveolar wall was thickened and there were exudate and inflammatory cell infiltration in the alveolar cavity.Compared with the model group,the amount of leukocyte,erythrocyte,blood platelet and hemoglobin and so on of middle dose and high dose of Dabupi Decoction group was increased(P<0.05).And the level of TGF-β1,IL-1β,IL-6,TNF-αand IL-8 of every Dabupi Decoction groups decreased significantly(P<0.05).And thymus index and spleen index also increased significantly after radiation(P<0.05).The pathological changes of right lung tissue of the rats improved.Conclusion:Dabupi Decoction can protect radiationinduced pulmonary injury by prohibiting the expression of TGF-β1,IL-1β,IL-6,TNF-αand IL-8 in serum.
