摘要:目的通过观察骨折不同分期治疗对SD大鼠骨折愈合特异性转录因子(Osx)表达的影响,为其能促进成骨细胞分化,促进骨折愈合提供一定的理论依据。方法选用SD大鼠40只,于双侧胫骨中下1/3段造成3 mm骨质缺损,随机分为生理盐水组、早期治疗组、中期治疗组、后期治疗组,每组10只,在相应时间段给予相应治疗,于造模后30 d处死大鼠,取胫骨标本,运用免疫组化法测定其Osx表达,并对相应的平均光密度值(MOD)进行分析。结果早、中、后三期治疗组对Osx表达均有促进作用,较生理盐水组高,差异有统计意义(P<0.05或P<0.01);各治疗组之间比较,后期治疗组对Osx的促进作用更显著(P<0.05),中期治疗组对Osx的促进作用亦优于早期治疗组(P<0.05)。结论中医辨证分期治疗骨折是必要的。
Objective To provide certain theoretical evidence for the functions of three-phase Chinese medicine treatment in promoting the osteoblast differentiation and fi'acture healing by observing its influences on the specific transcription factor (Osx) expression in fracture healing SD rats. Methods Chose 40 SD rats, make 3 mm bone defects in the lower 1/3 section of both of the tibias, and randomly divide the rats into four groups: saline groups, early stage treatment group, middle stage treatment group, later stage treatment group, 10 rats in each group. Apply corresponding treatment in certain stages, and killed the rats in the 30th day to take the specimen of the tibias. Determined the Osx expression with immunohistochemical methods and analysed the corresponding mean optical density (MOD) value. Results The promotion effects to Osx expression were showed in all the early, middle and later stage treatment groups, all of which were significantly higher than normal saline groups (P〈0.05 or P〈0.01 ) ; comparison among the three treatment groups showed that the promotion effects to Osx expression in the later stage treatment group was significantly higher than the other two groups (P〈0.05), and that of the middle stage treatment group was higher than the early stage treatment group (P〈0.05). Conclusion Chinese medicine treatments according to different pattens in different stages are necessary in bone fractures.
