摘要:目的基于免疫组化技术观察秦艽不同配伍对风寒湿痹类风湿关节炎(RA)大鼠踝关节基质金属蛋白酶-3(MMP-3)、基质金属蛋白酶组织抑制剂-1(TIMP-1)的影响。方法按随机数字表法将大鼠分为8组:空白组、Ⅱ型胶原模型组、风寒湿病证模型组、对照组、秦艽组、秦艽-威灵仙组、秦艽-桑寄生组、秦艽-防己组,每组10只。造模结束后,各中药组按25 g·kg^(-1)给予相应药液,对照组按6 mg·kg^(-1)给予雷公藤多苷混悬液,其余组给予等量0. 9%NaCl,1天1次,连续21 d。以免疫组化检测踝关节MMP-3、TIMP-1的表达。结果空白组、Ⅱ型胶原模型组、风寒湿病证模型组、对照组、秦艽组、秦艽-威灵仙组、秦艽-桑寄生组、秦艽-防己组的MMP-3光密度值分别为(3. 71±0. 80)×10~3,(9. 63±0. 18)×10~3,(11. 66±0. 62)×10~3,(6. 06±0. 94)×10~3,(7. 67±0. 46)×10~3,(5. 84±0. 39)×10~3,(7. 38±0. 37)×10~3,(8. 49±0. 37)×10~3;这8组的TIMP-1光密度值分别为(26. 62±3. 35)×10~3,(5. 64±1. 03)×10~3,(3. 94±0. 47)×10~3,(21. 77±2. 48)×10~3,(13. 80±3. 26)×10~3,(19. 77±2. 08)×10~3,(13. 48±2. 38)×10~3,(9. 14±1. 19)×10~3,Ⅱ型胶原模型组和病证模型组与空白组比较,MMP-3与TIMP-1表达的差异均有统计学意义(均P <0. 01);各给药组与病证模型组比较,MMP-3与TIMP-1表达差异均有统计学意义(均P <0. 01)。结论对于风寒湿痹型RA,平温相配的治疗效果优于平平相配、平寒相配及单味秦艽,实验结果与中医"寒者热之"的治则基本相符。
Objective To explore the influence of different compatibility of Gentianae Macrophyllae Radix( Qinjiao) on the expression of matrix metalloproteinase-3( MMP-3),tissue inhibitor of metalloproteinase-1( TIMP-1) in ankle joint of rheumatoid arthritis( RA) model rats. Methods SD rats were randomly divided into 8 groups: blank group,type Ⅱ collagen model group,wind-cold-dampness syndrome model group,control group,Qinjiao group,Qinjiao-Weilingxian group,Qinjiao-Sangjisheng group,Qinjiao-Fangji group. Type Ⅱ collagen model group was injected with type Ⅱ collagen emulsion only,except for the blank group; the other groups were injected with type Ⅱ collagenemulsion and given wind-cold-dampness stimulus to prepare the wind-cold-dampness RA rat model. After the establishment of the model,the blank group,the 2 model groups were given normal saline; the control group was given 6 mg·kg^-1 tripterygium glycosides suspension; Qinjiao group,Qinjiao-Weilingxian group,Qinjiao-Sangjisheng group,Qinjiao-Fangji Group were given respectively 25 g·kg^-1 corresponding medicinal liquid. The expression of MMP-3 and TIMP-1 in ankle joint was detected by immunohistochemistry. Results The optical density values of MMP-3 in blank group,type Ⅱ collagen model group,wind-cold-dampness syndrome model group,control group,Qinjiao group,Qinjiao-Weilingxian group,Qinjiao-Sangjisheng group,Qinjiao-Fangji group were respectively( 3. 71 ± 0. 80) × 10^3,( 9. 63 ± 0. 18) × 10^3,( 11. 66 ± 0. 62) × 10^3,( 6. 06 ± 0. 94) × 10^3,( 7. 67 ± 0. 46) × 10^3,( 5. 84 ± 0. 39) × 10^3,( 7. 38 ± 0. 37) × 10^3,( 8. 49 ± 0. 37) × 10^3; TIMP-1 optical density values in the 8 groups were( 26. 62 ± 3. 35) × 10^3,( 5. 64 ± 1. 03) × 10^3,( 3. 94 ± 0. 47) × 10^3,( 21. 77 ± 2. 48) × 10^3,( 13. 80 ± 3. 26) × 10^3,( 19. 77 ± 2. 08) × 10^3,( 13. 48 ± 2. 38) × 10^3,( 9. 14 ± 1. 19) × 10^3. Comparison between the 2 model groups and blank group,the difference had statistical significance( all P 0. 01); the difference between the wind-cold-dampness syndrome model group and the each administration groups had statistical significance( all P 0. 01). Conclusion For rheumatoid arthritis of wind-cold-dampness arthralgia type,the therapeutic effect of the combination of mild and warm is better than that of the combination of mild and the combination of cold and mild and single-taste Qinjiao. The experimental results are basically consistent with the therapeutic principle of"treating cold diseases should use hot medicine".
