摘要:目的:探讨镇肝熄风汤对自发性高血压大鼠(SHR)肠道紧密连接蛋白的影响。方法:50只SHR大鼠随机分为模型组,贝那普利组,中药高、中、低剂量组,10只WKY大鼠为空白组。中药低、中、高剂量组每日分别给予镇肝熄风汤60、30、15g·kg^(-1)·d^(-1)灌胃,贝那普利组给予盐酸贝那普利0.90mg·kg^(-1)·d^(-1)灌胃。空白组和模型组每日给予等体积蒸馏水灌胃,干预8周后测定各组大鼠血压,应用酶联免疫实验法检测各组大鼠血清二胺氧化酶(DAO)及D-乳酸的水平,采用免疫荧光法检测结肠组织紧密连接蛋白ZO-1、Occludin的变化。RT-PCR法检测结肠组织ZO-1、Occludin mRNA水平。结果:与模型组比较,各给药组收缩压、舒张压和平均动脉压显著降低(P<0.05),血清中DAO及D-乳酸均降低(P<0.01),各给药组免疫荧光ZO-1和Occludin蛋白的染色强度增强、ZO-1、Occludin mRNA在结肠中的表达显著升高(P<0.05)。结论:镇肝熄风汤可能通过影响紧密连接蛋白的表达,改善肠黏膜功能,从而发挥保护大鼠肠黏膜屏障,下调血压的作用。
Objective:To explore the influencing mechanism of Zhengan Xifeng Decoction on the expression of tight junction proteins in SHR.Methods:Fifty SHR rats were randomly divided into the model group,Benner Pury group,Zhengan Xifeng Decoction high,middle,and low dose groups.Ten WKY as the normal control group.Zhengan Xifeng Decoction high,medium,and low dose groups were fed with Zhengan Xifeng Decoction 60,30,15 g·kg^-1·d^-1,respectively,Benazepril group were fed with Benazepril 0.90 mg·kg^-1·d^-1.The normal control group and model group were fed with distilled water daily.After the intervention period of 8 weeks,the blood pressure,the level of DAO,and D-lactate level in serum was detected,the expression of ZO-1,Occludin,and its mRNA in the colon was detected by immunofluorescence and RT-PCR.Results:Compared with the model group,the systolic pressure,diastolic pressure,and mean arterial pressure of treatment groups were significantly reduced(P<0.05).DAO and D-lactate levels in treatment groups significantly reduced(P<0.01);the expression of ZO-1,Occludin mRNA in the colon was higher in the treatment group.Conclusion:Zhengan Xifeng Decoction can protect the intestinal mucosal barrier by regulating the expression of tight junction proteins and reduce blood pressure in SHR.
