摘要:目的:比较参苓白术散与痛泻要方对三硝基苯磺酸(TNBS)/乙醇法建立的溃疡性结肠炎模型大鼠骨髓与外周血来源骨髓间充质干细胞(BMSCs)生长及增殖的干预作用,同时"以方测证"探讨该动物模型与脾虚湿盛和肝郁脾虚证是否相关。方法:120只雄性SD大鼠随机分为空白组、模型组、参苓白术散组、痛泻要方组,每组30只,TNBS/乙醇法建立溃疡性结肠炎大鼠模型,给予参苓白术散与痛泻要方灌胃治疗,分离外周血与骨髓中BMSCs并进行培养,流式细胞技术(FCM)检测BMSCs生长周期,实时无标记细胞分析技术(RTCA)检测BMSCs增殖能力。结果:参苓白术散组与痛泻要方组具有降低骨髓及外周血来源BMSCs的G1期细胞比例(P<0.05),升高S期细胞比例(P<0.05),且痛泻要方组优于参苓白术散组(P<0.05);模型组外周血来源BMSCs的增殖能力被抑制,参苓白术散组与痛泻要方组具有促进外周血来源BMSCs增殖作用(P<0.05),痛泻要方组优于参苓白术散组(P<0.05),而二方促进骨髓来源BMSCs增殖作用无差异。结论:参苓白术散与痛泻要方能促进骨髓与外周血来源BMSCs生长与增殖,从而确保足够数量的BMSCs归巢到结肠黏膜发挥修复作用,这可能是其治疗溃疡性性结肠炎的作用机制之一;TNBS乙醇法建立UC大鼠模型可能为中医肝郁脾虚证。
Objective:To compare the intervention effects of Shenling Baizhu Powder and Tongxie Yaofang on the growth and proliferation of BMSCs derived from bone marrow and proliferation blood in rats with ulcerative colitis induced by TNBS/rthanol,and to explore whether the animal model is related to spleen deficiency with dampness or liver depression and spleen deficiency.Methods:A total of 120 SD male rats were randomly divided into control group,model group,Shenling Baizhu Powder group and Tongxie Yaofang group,30 rats in each group.Rats’ model of ulcerative colitis was established by TNBS/ethanol method.The rats were given Shenling Baizhu Powder and Tongxie Yaofang by gastric lavage.BMSCs in peripheral blood and bone marrow were isolated and cultured.the growth cycle of BMSCs was detected by flow cytometry (FCM),and the proliferation of BMSCs was detected by real-time labeless cell analysis.Results:Shenling Baizhu Powder group and Tongxie Yaofang group could decrease G1 phase cell ratio of BMSCs from bone marrow and peripheral blood (P<0.05),increased S phase cell ratio (P<0.05),and Tongxie Yaofang group was superior to Shenling Baizhu Powder group (P<0.05).The proliferation ability of BMSCs from peripheral blood in model group was inhibited,and the Shenling Baizhu Powder and Tongxie Yaofang can promote the proliferation of BMSCs from peripheral blood (P<0.05),Tongxie Yaofang group was better than Shenling Baizhu Powder group (P<0.05),but the two recipes have no difference in promoting the proliferation of BMSCs from bone marrow.Conclusion:Shenling Baizhu Powder and Tongxie Yaofang can promote the growth and proliferation of BMSCs derived from bone marrow and peripheral blood,so as to ensure that enough BMSCs homing to the colonic mucosa to play a repair role,which may be one of the mechanisms of its treatment of ulcerative colitis,furthermore,the establishment of UC rat model by TNBS ethanol may be the syndrome of liver depression and spleen deficiency.
