摘要:目的探讨当归红芪超滤物抗辐射致乳鼠心肌细胞损伤的作用及其可能机制。方法用X线照射Wistar乳鼠原代培养的心肌细胞建立辐射损伤模型,用不同浓度的当归红芪超滤物进行干预,实验分正常对照组、辐射损伤模型组、当归红芪超滤物不同浓度(3、6、9 mg·mL-1)干预组(UFE-AH)。MTT法检测各组心肌细胞的存活率,2,4二硝基苯肼显色法检测各组细胞培养上清液中乳酸脱氢酶(LDH)的含量,黄嘌呤氧化酶法测定各组细胞内超氧化物歧化酶(SOD)的活性,免疫组化法观测各组细胞Bax的表达,蛋白印迹半定量测定各组细胞Bax蛋白的表达。结果与辐射损伤模型组比较,当归红芪超滤物各干预组心肌细胞存活率明显升高,LDH含量显著降低(P<0.05),SOD活性显著提高(P<0.05),Bax表达显著降低(P<0.05)。结论当归红芪超滤物具有拮抗辐射致心肌细胞损伤的作用,其机制与清除自由基有关。
Objective To explore the effect and potential mechanism on myocardial cellular damage induced by radiation in suckling mice treated with ultra- infiltration extract from Angelica Sinensis and Hedysarum Polybotrys mixture. Methods X- ray irradiation was used on the primitively cultured myocardial cells of Wistar suckling mice to set up radiation damage models. The ultra- infiltration extract from Angelica Sinensis and Hedysarum Polybotrys mixture was used for intervention at different concentrations. In the experiment,a normal control group,a radiation damage model group,and the intervention groups of different concentrations( 3,6,9 mg·mL^-1 of the ultra- infiltration extract from Angelica Sinensis and Hedysarum Polybotrys mixture) were divided. MTT method was used to detect the survival rate of myocardial cells in each group. The 2,4 dinitro- phenylhydrazine colorimetric reaction was used to detect LDH content in the culture supernatant. The xanthin oxidase enzyme method was used to detect SOD activity. IHC method was used to detect Bax expression and Western blotting was to detect Bax protein expression in each group. Results Compared with the radiation damage model group,the survival rate of myocardial cell was increased apparently in each intervention group,LDH content was reduced significantly( P 0. 05),SOD activity was increased significantly( P 0. 05) and Bax expression was reduced significantly( P 0. 05). Conclusion The ultra-infiltration extract from Angelica Sinensis and Hedysarum Polybotrys mixture resists on myocardial cellular damage induced by radiation and its mechanism is related to the clearance of free radicals.
