摘要:目的:捕获当归治疗痛经活性组分富集分离片段。方法:采用硅胶柱色谱,用乙酸乙酯∶石油醚1∶9,乙酸乙酯:石油醚4∶6,乙酸乙酯,甲醇依次洗脱,得到流分1,流分2,流分3,流分4洗脱片段,片段组分差异用HPLC指纹图谱表征;采用离体实验观察分离片段干预缩宫素诱导的子宫平滑肌、正常子宫平滑肌的生物效应。结果:与空白组比较,20mg/L流分1、80mg/L流分2有显著的抑制缩宫素引发的子宫平滑肌收缩;与20mg/L流分1比较,160mg/L流分3或流分4抑制缩宫素诱导的子宫平滑肌收缩差异无显著性;与对照组比较,40mg/L流分1,160mg/L流分2松弛正常子宫平滑肌的作用显著;与10mg/L流分1比较,160mg/L流分3或流分4松弛正常子宫平滑肌作用差异无显著性,显示流分3或流分4直接松弛子宫平滑肌作用微弱。结论:当归中低极性物质是治疗痛经的活性组分,其主要富集在流分1、流分2。
Objective :To capture separated fraction enriched in bioactive constituent of relief dysmenorrhea. Methods: Chromatographic methods were used to obtain the different separated fraction, the petroleum ether extract was subjected to open column chromatography on normal phase silica gel and respectively eluted with petroleum ether/ethyl acetate (9:1 v/v), petroleum ether/ethyl acetate (6:4 v/v) , ethyl acetate, and finally eluted with methanol, totally obtained were 4 fractions, which were respectively Fraction!, Fraction2, Fraction3, Fraction4. Their components difference are characterized by HPLC fingerprint. Each fraction was investigated to suppress spontaneous contractions and oxyto- cin induced contractions of rat uterine smooth muscle in vitro. The data is expressed as the mean + SD, Statistical significance of differences among groups was assessed using one-way ANOVA by SPSS11.0. Results:The results show that 20mg/L Fractionl or 80mg/L Fraction2 be- gin to suppress oxytocin induced contractions of rat uterine smooth muscle in vitro, but Fraction3 160mg/L or Fraction3 160mg/L suppression oxytocin induced contractions is equal with Fractionl 20mg/L, no significance of differences between Fraction3 or Fraction4 160mg/L and Fractionl 20mg/L. Fractionl 40mg/L and Fraction2 160mg/L can relax rat uterine smooth muscle of loading 2 g. Fraction3 160mg/Land Fraction3 160mg/L relax rat uterine smooth muscle of loading 2 g is equal with Fractionl 10mg/L, the result shows that Fraction3 and Frac- tion4 have weaker relaxation on rat uterine smooth muscle. Conclusion: Fractionl and Fraction2 are fractions enriched in rehef-dysmenorrhe- al-bioactive-constituents that are low polar components of Angeheae sinensis root.
