摘要:目的:研究探讨敦煌医方瞿麦汤对肾草酸钙结石模型鼠Nrf2-NQO1通路的影响。方法:SD雄性大鼠60只按随机数字表法分为空白对照组(等体积蒸馏水)、模型组(等体积蒸馏水)、肾石通颗粒组(5 g/kg·d),敦煌医方瞿麦汤高、中、低剂量组(20、10、5 g/kg)每组各10只。大鼠肾草酸钙结石模型的制备采用1%乙二醇+2%氯化铵的溶液联合灌胃法,造模5 h后给予相应药物干预,持续4周。末次给药24 h后,检测各组大鼠的体质量、肾指数、血清钙(calcium,Ca^2+)、磷(phosphorus,P)含量、肾组织SOD活性、MDA含量、Nrf2 mRNA和QNO1mRNA的表达。结果:与空白对照组比较,模型组大鼠肾指数、血清Ca^2+含量、肾组织MDA的含量均增加,其体质量、肾组织SOD活性、Nrf2mRNA和NQO1mRNA的表达均下降;与模型组比较,敦煌医方瞿麦汤各干预组肾指数、血清Ca^2+含量均下降,体质量和肾组织SOD活性均升高,MDA含量均下降;敦煌医方瞿麦汤中、高剂量组肾组织Nrf2 mRNA和NQO1mRNA表达均升高。结论:敦煌医方瞿麦汤可通过上调Nrf2-NQO1通路相关因子表达,抑制大鼠肾草酸钙结石的形成。
Objective To study the Nrf2-NQO1 pathway effects of Dunhuang Yifang Qu Mai decoction on calcium oxalate nephrolithiasis rats.Method 60 male SD rats were randomly divided into blank control(equal volume distilled water)group,model(equal volume distilled water)group,Shenshitong granules(5 g/kg)group,Dunhuang Yifang Qu Mai decoction high-dose,middle-dose and low-dose(20、10、5 g/kg.d)groups,10 for each group.Except for blank control group,all the groups were given ig 1% ethylene glycol and 2% ammonium chlorid(2% NH4Cl)to establish calcium oxalate nephrolithiasis model.after 5 h of model establishment,corresponding drugs were given ig for 4 weeks.After 24 h of the last treatment,the weight body and the kidney index were monitored,the contents of serum Ca^2+,P were detected,and the levels of kidney SOD,MDA,Nrf2 and NQO1 were determined.Result Compared with those of blank control group,the kidney index,content of serum Ca^2+,MDA content in kidney tissue were increased and the weight body,SOD antivity,Nrf2 and NQO1mRNA expressions were all lowed in model group,with significant differences(P<0.05).Compared with those of model group,the kidney indexes were markedly decreased in Dunhuang Yifang Qu Mai decoction intervention group,the contents of serum Ca^2+ and MDA contents in kidney tissue were both decreased obviously in Dunhuang Yifang Qu Mai decoction intervention group,the weight body and the SOD antivities in kidney tissue were obviously increased in Dunhuang Yifang Qu Mai decoction intervention group,the Nrf2 and NQO1mRNA expressions were both raised sharply in Dunhuang Yifang Qu Mai decoction middle-dose and high-dose group.Conclusion Dunhuang Yifang Qu Mai decoction could effectively inhibit the formation of calcium oxalate stone in rats via increasing the relevent cytokines expressions of Nrf2-NQO1 pathways.
