摘要:目的比较植物雌激素白藜芦醇和选择性雌激素受体调节剂他莫昔芬对雌二醇诱发的去卵巢SD大鼠子宫内膜渗出和对血清VEGF浓度的影响。方法 30只SD大鼠随机分为5组:假手术组(Sham)、卵巢切除组(Ovx)、卵巢切除+17β-雌二醇组(Ovx+Est)、卵巢切除+17-β雌二醇+他莫昔芬组(Ovx+Est+Tam)、卵巢切除+17-β雌二醇+白藜芦醇组(Ovx+Est+Res)各6只,给药21 d后取材。用光学显微镜对各组子宫内膜形态学变化进行观察、对嗜酸性粒细胞数目进行计数,并用酶联免疫分析(ELISA)法测出各组血清VEGF的浓度。结果大鼠卵巢切除后,子宫明显萎缩且重量下降。给予外源性雌激素后,子宫重量及子宫系数增加;子宫被覆上皮细胞和腺体增生明显,子宫内膜中有嗜酸性粒细胞的渗出。Ovx+Est+Tam组和Ovx+Est+Res组血清VEGF浓度低于Ovx+Est组(P<0.05);Ovx+Est+Tam组渗出明显少于Ovx+Est+Res组(P<0.05)。结论白藜芦醇和他莫昔芬能发挥雌激素拮抗作用,使子宫内膜增生,能使雌二醇诱发的去卵巢大鼠子宫内膜渗出和血清VEGF浓度下降。
[ Objective ] To compare the effects of Resveratrol and Tammixifen on endometrial exudates and the concentrations of serum VEGF that induced by Estrogen in ovariectomized rats. [Methods] 30 rats model of SD randomly divided into 5 groups: Sham; Ovx; Ovx + Est; Ovx + Est + Tam; Ovx + Est + Res, using ELISA method to observe the changes of the concentration of VEGF in the serum, using optical microscope to invite endometrial mor- phological changes and count the Eosinophilic granulocyte, after delivered 21 days. [Results] It is found out that the weight of the ovariectomized Rats uterus decreased and the uterus is significantly atrophic after operation. While the weight and coefficient of the uterus is increased; and using microscope we demonstrated that the uterine epithe- lial cells and glands are obviously hyperplastic and after the exogenous phytoestrogens are given. The concentrationof serum VEGF in Ovx+Est+Tam and Ovx+Est+Res group is significantly lower than that of Ovx+Est group. The en- dometrial exudates in Ovx+Est+Tam group is significantly lower than that of Ovx+Est and Ovx +Est+Res group. [ Conclusion ] Tamoxifen can make serum VEGF expression significantly decreased, and it is antagonistic to estra- diol, which has a role on endometrial exudate of ovariectomized rats. Resveratrol can play a weak estrogen-like function and estrogen antagonist effect, and it can make the endometrial oozy and serum VEGF concentration de- creased, but the effects are weaker than tamoxifen.
