摘要:目的 探讨湿疹膏对2,4-二硝基氯苯(DNCB)诱导的特应性皮炎(AD)大鼠Janus激酶1信号转导和转录激活因子6(JAK1/STAT6)信号通路干预作用并探讨其机制。方法 用外涂DNCB溶液建立AD大鼠模型。将大鼠随机分为空白组(不做治疗干预)、模型组(不做治疗干预)、阳性对照组(0.25 g·cm^(-2)丁酸氢化可的松乳膏)和低、中、高剂量实验组(0.125、0.25、0.50 g·cm^(-2)湿疹膏),每组10只。末次用药24 h后,用酶联免疫吸附试验法检测相关炎症因子水平,用免疫组织化学法检测通路相关磷酸化蛋白的相对表达水平。结果 空白组、模型组、阳性对照组和高、中、低剂量实验组的白细胞介素(IL)-4水平分别为(3.10±0.45)、(11.11±0.06)、(4.00±1.05)、(4.40±1.16)、(5.24±1.21)和(8.78±1.22) pg·mL^(-1),IL-5水平分别为(9.58±0.19)、(22.25±0.28)、(9.83±0.68)、(10.59±1.20)、(11.46±2.50)和(14.76±1.35) pg·mL^(-1),IL-13水平分别为(18.30±0.65)、(53.73±2.18)、(23.26±2.73)、(27.14±4.17)、(42.28±1.44)和(48.75±1.57) pg·mL^(-1),磷酸化Janus激酶(p-JAK)1蛋白相对表达水平分别为2.01±0.47、2.97±0.01、2.04±0.41、2.04±0.38、2.04±0.63和2.14±0.32,磷酸化信号转导与转录激活子(p-STAT)6蛋白相对表达水平分别为1.14±0.06、1.89±0.81、1.10±0.48、1.11±0.39、1.26±0.44和1.78±0.32。模型组的上述指标与高、中剂量实验组和空白组比较,在统计学上差异均有统计学意义(P<0.01,P<0.05)。结论 湿疹膏可以有效调节特应性皮炎大鼠组织中IL-4、p-JAK1、p-STAT6的蛋白表达,抑制IL-5、IL-13等与特应性皮炎相关的关键炎症因子,减少组织炎症损伤,其作用机制可能与干预JAK1-STAT6通路有关。
Objective To investigate the effect of eczema ointment on Janus kinase 1/signal transduction and activator of transcription 6(JAK1/STAT6)signaling pathway in 2,4-dinitrochlorobenzene(DNCB)-induced atopic dermatitis(AD)rats and explore its mechanism.Methods The rat model of AD was established by applying DNCB solution.The rats were divided into blank group(no intervention),model group(no intervention),positive control group(0.25 g·cm^(-2) hydrocortisone butyrate cream)and experimental-L,-M,-H groups(0.125,0.25,0.50 g·cm^(-2) eczema cream)according to random number table,with 10 rats in each group.24 h after the last administration,the levels of related inflammatory factors were detected by enzyme-related immunosorbent assay(ELISA),the relative expression levels of pathway-related phosphorylated proteins were detected by immunohistochemical assay(IHC).Results The IL-4 levels of blank group,model group,positive control group and experimental-H,-M,-L groups were(3.10±0.45),(11.11±0.06),(4.00±1.05),(4.40±1.16),(5.24±1.21)and(8.78±1.22)pg·mL^(-1);IL-5 levels were(9.58±0.19),(22.25±0.28),(9.83±0.68),(10.59±1.20),(11.46±2.50)and(14.76±1.35)pg·mL^(-1);IL-13 levels were(18.30±0.65),(53.73±2.18),(23.26±2.73),(27.14±4.17),(42.28±1.44)and(48.75±1.57)pg·mL^(-1);the relative expression levels of phosphorylated Janus kinase(p-JAK)1 were 2.01±0.47,2.97±0.01,2.04±0.41,2.04±0.38,2.04±0.63 and 2.14±0.32;the relative expression levels of phosphorylated signal transducter and activator of transcriptional(p-STAT)6 protein were 1.14±0.06,1.89±0.81,1.10±0.48,1.11±0.39,1.26±0.44 and 1.78±0.32,respectively.Compared with the model group,the above indexes in the blank group and experimental-M,-H groups were statistically significant(P<0.01,P<0.05).Conclusion The eczema ointment can effectively regulate the protein expressions of IL-4,p-JAK1,and p-STAT6 in the tissues of atopic dermatitis rats,inhibit key inflammatory factors such as IL-5 and IL-13 related to atopic dermatitis,reduce tissue inflammatory damage,and its mechanism of action may be related to the intervention of the JAK1-STAT6 pathway.
