摘要:目的了解河北地区一株人轮状病毒的基因组特征和进化关系。方法收集我国病毒性腹泻监测网络采集的5岁以下腹泻住院儿童粪便标本,分型发现一株G9P[6]型轮状病毒感染的样本。通过探针捕获结合高通量测序、RT-PCR的方法,获得了这株G9P[6]毒株全基因组序列并进行了基因组特征分析。结果分析结果显示该毒株属于G9-P[6]-I5-R1-C1-M1-A8-N1-T1-E1-H1基因型。核苷酸序列同源性和系统发育分析表明,该病毒株的VP4、VP2、VP1、NSP4基因与人源毒株亲缘关系最近,最高同源性为95.6%~98.5%,其余7个基因与多株猪源毒株的亲缘关系较近,但相近的毒株谱系并不相同。其中,VP3与Panda/CH-1株和Human/LL3354株同源性最高,并且与Panda/CH-1株、人和猪来源的多个病毒株聚在一起;VP7与Human/LL51695株、Dog/SCCD-A株和Pig/TM-a-P1株处于同一进化支。VP6和NSP2则与Dog/SCCD-A株和Pig/SCMY-A3株显示出最高同源性,并归于同一进化分支。NSP1与多株猪来源轮状病毒具有较高同源性,并与其聚为同一进化分支。NSP3和NSP5与多株人和猪的轮状病毒显示较高同源性,并聚在一起。结论该病毒株可能是由源自人类与猪的病毒株之间发生跨种属传播后,经过基因重配而形成的一种新型A组轮状病毒毒株。该11个节段的型别组合尚属首次报道,丰富了我国轮状病毒基因型。
ObjectiveTo investigate the genomic characteristics and evolutionary relationship of a human rotavirus strain from Hebei province.MethodsFecal samples of hospitalized children under 5 years old with diarrhea were collected through the surveillance network of viral diarrhea in China.One strain of G9P[6]type rotavirus was detected from one sample.The whole genome of this G9P[6]strain was obtained by using combined probe-capture and high-throughput sequencing method as well as RT-PCR and the genome characteristics were then analyzed.ResultsThe analysis result showed that this strain belonged to genotype G9-P[6]-I5-R1-C1-M1-A8-N1-T1-E1-H1.Nucleotide sequence homology and phylogenetic analysis showed that the VP4,VP2,VP1 and NSP4 genes were closely related to the human strain,with highest similarity of 95.6%-98.5%.The other 7 genes were closely related to several pig strains,but the genealogy of similar strains was different.Among them,VP3 had the highest similarity with Panda/CH-1 strain and Human/LL3354 strain,and was clustered with Panda/CH-1 strain and multiple virus strains derived from human and pig.The VP7 gene was phylogenetic clustered with Human/LL51695,Dog/SCCD-A and Pig/TM-a-P1.The VP6 and NSP2 gene showed the highest similarity to Dog/SCCD-A and Pig/SCMY-A3 strain,and were phylogentic grouped into one cluster.NSP1 has high homology with many strains of porcine rotavirus and is clustered into the same evolutionary clade.NSP3 and NSP5 showed high homology with multiple strains of human and porcine rotavirus and clustered together.ConclusionsIt is speculated that HB16 strain may be a novel group A rotavirus strain formed by genetic reassorting after cross-species transmission between human and porcine virus strains.This is the first report on the genetic combination of the 11 segments which enriched the rotavirus genotype in China.
