摘要:目的观察四神丸对脾肾阳虚型溃疡性结肠炎(UC)大鼠血清白细胞介素-8(IL-8)、肿瘤坏死因子-α(TNF-α)含量及结肠组织核因子κB(NF-κB) p65蛋白表达的影响,从而探讨四神丸治疗脾肾阳虚型溃疡性结肠炎的作用机制。方法采用随机数字表法将大鼠分为空白组,模型组,阳性对照组,四神丸高、中、低剂量组6组,采用"病-证结合"的方法制备脾肾阳虚型UC大鼠模型,各组给予相应药物灌胃。观察各组大鼠结肠黏膜损伤情况并评分,采用酶联免疫法(ELISA)和免疫组化SP法分别检测各组大鼠血清IL-8、TNF-α的含量及结肠组织NF-κB p65蛋白的表达水平。结果与空白组比较,模型组大鼠结肠黏膜损伤评分,血清IL-8、TNF-α含量均明显升高,结肠组织NF-κB p65蛋白表达明显增强,差异均有统计学意义(P<0.01);与模型组比较,各治疗组(阳性对照组及四神丸高、中、低剂量组)大鼠结肠黏膜损伤评分,血清IL-8、TNF-α含量均明显降低,阳性对照组及四神丸中、高剂量组结肠组织NF-κB p65蛋白表达明显下调,差异均有统计学意义(P<0.05或P<0.01)。结论四神丸可明显降低脾肾阳虚型UC模型大鼠血清IL-8、TNF-α的含量,下调结肠组织NF-κB p65蛋白的表达,改善结肠黏膜损伤。
Objective To observe the effects of Sishen Wan(四神丸) on the serum levels of IL-8,TNF-α and expression of colonic NF-κB p65 protein in ulcerative colitis( UC) rats with deficiency of spleen and kidney yang type,and to explore the mechanism of Sishen Wan in the treatment of UC with deficiency of spleen and kidney yang type. Methods The experimental rats were randomly divided into 6 groups according to random number table,including blank group,model group,positive control group,and the high,medium and low dose Sishen Wan groups,using "combination of disease and syndrome method"for the preparation of UC rats model with deficiency of spleen and kidney yang type,each group was given the corresponding medicine gavage. The colonic mucosal injury of the rats in each group was observed and scored.The serum levels of IL-8,TNF-α and expression of NF-κB p65 protein in colonic tissue of rats in each group was detected by enzyme-linked immunosorbent assay( ELISA) and immunohistochemical SP method. Results The colonic mucosal injury score,the serum levels of IL-8 and TNF-α in the model group were significantly higher than those in the blank group,there were statistically significant differences between the two groups( P<0.01);but compared with the model group,the colonic mucosal injury score,the serum levels of IL-8,TNF-α of each treatment group( the positive control group,and the high,medium and low dose Sishen Wan groups) decreased significantly. The expression of NF-κB p65 protein in colonic tissue down regulated significantly in the positive control group and the high,medium dose Sishen Wan groups,there were statistically significant differences( P<0.05 or P<0.01). Conclusion Sishen Wan can significantly decrease the serum levels of IL-8,TNF-α in UC rats model with deficiency of spleen and kidney yang type,down regulate the expression of NF-κB p65 protein in colon tissue,and improve the colonic mucosa injury.
