摘要:目的通过测定岷县当归与临洮当归中阿魏酸、阿魏酸松柏酯、藁本内酯、洋川芎内酯A、洋川芎内酯I、洋川芎内酯H、正丁基苯酞、欧当归内酯等8种有效成分的含量,对比评价新产区临洮所产当归的药材质量。方法采用高效液相色谱(HPLC)法测定岷县当归与临洮当归中以上8种有效成分的含量,色谱柱为Merk RP-C18柱(250.0 mm×4.6 mm,5μm),流动相为乙腈(B)-1%乙酸(A),梯度洗脱,检测波长280 nm,柱温30℃,流速1 m L/min,进样量20μL;采用SPSS 21.0统计软件比较2个产地8种有效成分的含量差异。结果该色谱条件可以将当归中以上8种有效成分完全分离并同时测定其含量;8种有效成分在各自线性范围内均呈现良好的线性关系;2个产地当归中有效成分除洋川芎内酯I与洋川芎内酯H外,其余6种有效成分含量差异均有统计学意义(P<0.05)。结论该方法简单可行、重复性好,可为临洮合理引种当归及其规范化栽培提供参考。
Objective To conduct a comparative evaluation on the medicinal material quality of Angelica sinensis from Lintao,a new producing area,by content determination of 8 active principles of Angelica sinensis between Minxian County and Lintao County,including ferulic acid,coniferyl ferulate,ligustilide,senkyunoli A,senkyunoli I,senkyunoli H,n-butylphthalide and lebistilide A.Methods High performance liquid chromatography(HPLC)was used to content determine the 8 active principles in Angelica sinensis between Lintao County and Minxian County.Merk RP-C18 column(250.0 mm×4.6 mm,5μm)and acetonitrile as the mobile phase B and 1%acetic acid as mobile phase A for gradient elution.The detection wavelength was 280 nm,column temperature was 30℃and flow rate was 1 m L/min,and the injected amount was 20μL.The content differences of 8 active principles from 2 producing areas were compared by SPSS 21.0 software.Results The above 8 active principles in Angelica sinensis could be completely separated and their content could be determined simultaneously by the chromatographic conditions.The 8 active principles showed a good linear relationship in their respective linear ranges;the content of the other 6 active principles in Angelica sinensis from the 2 producing areas were statistically significant(P<0.05),except for senkyunoli I and senkyunoli H.Conclusion The method is simple,feasible and repeatable,which can provide a reference for the reasonable introduction of Angelica sinensis and its standardized cultivation.
