摘要:目的探讨“二龙戏珠”针法对动眼神经麻痹(ONP)大鼠眼外肌功能及动眼神经半乳糖凝集素-1(Galectin-1)mRNA表达的影响。方法50只大鼠随机分为空白组(BG)、假手术组(Sham)、模型组(MG)、电针组(EA)、二龙戏珠针法组(ELXZ),BG组大鼠不进行处理,Sham组仅暴露动眼神经,MG组、EA组、ELXZ组暴露并夹闭动眼神经建立ONP模型。造模成功后,EA组大鼠针刺眼上直肌、下直肌并使用电针仪干预;ELXZ组行“二龙戏珠”针法针刺太阳穴,每日1次,每次10 min,6 d为1个疗程,连续干预4个疗程。造模完成后(干预前)和干预后采用数字化图像分析技术测量大鼠瞳孔直径和睑裂高度,肌张力换能器检测大鼠眼外肌肌肉张力,实时荧光定量PCR(qRT-PCR)检测大鼠动眼神经Galectin-1 mRNA表达。结果(1)瞳孔直径:EA组、ELXZ组ONP大鼠干预后瞳孔直径较干预前变短,MG组大鼠瞳孔直径长于BG组和Sham组,而EA组、ELXZ组短于MG组,差异均有统计学意义(均P<0.05)。(2)睑裂高度:EA组、ELXZ组ONP大鼠干预后睑裂高度较干预前变大,MG组大鼠睑裂高度小于BG组和Sham组,而EA、ELXZ组大于MG组,ELXZ组大于EA组,差异均有统计学意义(均P<0.05)。(3)肌张力:EA组、ELXZ组ONP大鼠干预后眼外肌肌张力较干预前提高,MG组大鼠眼外肌的肌张力小于BG组和Sham组,差异均有统计学意义(均P<0.05)。EA组大鼠眼外肌的肌张力与MG组比较,差异无统计学意义(P>0.05)。ELXZ组大鼠眼外肌的肌张力大于MG组和EA组,差异均有统计学意义(均P<0.05)。(4)Galectin-1 mRNA:MG组大鼠动眼神经Galectin-1mRNA表达低于BG组和Sham组,EA组、ELXZ组高于MG组,差异均有统计学意义(均P<0.05)。而EA组与ELXZ组大鼠比较,差异无统计学意义(P>0.05)。结论“二龙戏珠”针法可以改善ONP模型大鼠眼外肌肌张力、缩小瞳孔、扩大睑裂,增加动眼神经中Galectin-1mRNA的表达。
OBJECTIVE To investigate the effects of the Erlong Xizhu acupuncture technique on extraocular muscle function and oculomotor nerve Galectin-1 mRNA expression in rats with oculomotor nerve palsy(ONP).METHODS A total of 50 rats were randomly assigned to the blank group(BG),sham-group(Sham),model group(MG),electroacupuncture group(EA),and Erlong Xizhu acupuncture group(ELXZ).The rats in the BG group received no intervention;The rats in the Sham group underwent surgical exposure of the oculomotor nerve without injury;The rats in the MG,EA,and ELXZ groups underwent exposure and clamping of the oculomotor nerve to establish the ONP model.Following successful modeling,the EA group received acupuncture at the superior rectus and inferior rectus muscles with electrical stimulation;The ELXZ group received Erlong Xizhu acupuncture once daily for 10 mins,six days as one course,for four consecutive courses.Before and after intervention,digital image analysis was used to measure pupil diameter and palpebral fissure height;a muscle tension transducer was employed to assess extraocular muscle tension;And quantitative real-time PCR(qRT-PCR)was performed to determine oculomotor nerve Galectin-1 mRNA expression.RESULTS(1)Pupil diameter:After intervention,pupil diameter decreased in EA and ELXZ groups compared with pre-intervention values.The MG group showed significantly larger pupil diameters than the BG and Sham groups,while EA and ELXZ groups had smaller pupil diameters than the MG group.The differences were all statistically significant(P<0.05).(2)Palpebral fissure height:After intervention,palpebral fissure height increased in EA and ELXZ groups.The MG group exhibited significantly reduced palpebral fissure height compared with BG and Sham groups,whereas EA and ELXZ groups had greater values than the MG group,with ELXZ higher than EA.The differences were all statistically significant(P<0.05).(3)Muscle tension:After intervention,extraocular muscle tension was elevated in EA and ELXZ groups compared with pre-intervention.The MG group had significantly lower muscle tension than the BG and Sham groups.The differences were all statistically significant(P<0.05).No significant difference was observed between EA and MG groups(P>0.05),whereas ELXZ showed significantly higher muscle tension than both MG and EA groups.The differences were all statistically significant(P<0.05).(4)Galectin-1 mRNA expression:The MG group had significantly reduced oculomotor nerve Galectin-1 mRNA expression compared with BG and Sham groups.Both EA and ELXZ groups exhibited significantly higher expression than the MG group.The differences were all statistically significant(P<0.05).No significant difference between EA and ELXZ groups(P>0.05).CONCLUSIONS The Erlong Xizhu acupuncture technique can improve extraocular muscle tension,reduce pupil diameter,enlarge palpebral fissure height,and increase Galectin-1 mRNA expression in the oculomotor nerve of ONP model rats.
