摘要:为了探究不同层积处理对羌活(Notopterygium incisum Ting ex H.T.Chang)种子发芽率的影响,以及层积过程中种子的生理生化变化规律,将羌活种子进行室内不同层积基质处理和野外原位层积,对经过层积处理的羌活种子,测定其种胚形态、发芽率以及生理生化指标。结果表明,随着层积时间的延长,羌活种胚逐渐发育为子叶形胚。羌活种子经过变温层积(20℃/4℃90 d+4℃120 d),种子休眠解除;室内层积处理中,A处理(河沙)发芽率最高(53.49%),B处理(V_(河沙)∶V_(蛭石)=1∶1)次之,C处理(V_(河沙)∶V_(珍珠岩)=1∶1)最低;野外原位层积7个月,发芽率显著高于室内不同层积基质处理,发芽率为60.43%。羌活种子层积过程中,淀粉含量显著降低,蛋白质含量先升高后下降,可溶性糖含量先下降后升高;过氧化物酶(POD)和超氧化物歧化酶(SOD)活性在种子层积过程中显著变化,丙酮酸激酶(PK)活性显著升高;内源激素赤霉素(GA_(3))和吲哚乙酸(IAA)含量显著升高,而脱落酸(ABA)含量显著下降。胚率与淀粉含量极显著负相关,与丙酮酸激酶活性极显著正相关,与IAA含量显著正相关。羌活种子休眠解除须满足暖/冷交替-低温的变化,层积过程中种子内部贮藏营养物质含量降低、酶活性显著变化,ABA含量降低以及GA_(3)和IAA含量升高,协同作用促进羌活种子休眠解除。
In order to explore the germination rate of Notopterygium incisum seeds under different stratification treatments and the physiological and biochemical changes of the seeds during the stratification process,the Notopterygium incisum seeds were treated with different stratification substrates indoors and in situ in the field.The embryo morphology,germination rate and physiological and biochemical indicators of the Notopterygium incisum seeds after stratification treatment were determined.The results indicated that with the extension of stratification time,the seed embryos of Notopterygium incisum gradually developed into cotyledonoid embryos.The seeds of Notopterygium incisum were released from dormancy after variable temperature stratification(20℃/4℃for 90 days+4℃for 120 days).Among the indoor stratification treatments,the germination rate of river sand stratification was the highest,at 53.49%,followed by V river sand∶V vermiculite=1∶1,and the lowest was V river sand∶V perlite=1∶1.The germination rate of in-situ layering in the field for 7 months was significantly higher than that of different layering matrix treatments indoors,with a germination rate of 60.43%.During the stratification process of Notopterygium incisum seeds,the starch content decreased significantly,the protein content increased first and then decreased,and the soluble sugar content decreased first and then increased.The activities of peroxidase(POD)and superoxide dismutase(SOD)changed significantly during the seed stratification process,and the activity of pyruvate kinase(PK)increased significantly.The contents of endogenous hormones gibberellin(GA 3)and indoleacetic acid(IAA)significantly increased,while the content of abscisic acid(ABA)significantly decreased.Embryo rate was extremely significantly negatively correlated with starch content,extremely significantly positively correlated with pyruvate kinase activity,and significantly positively correlated with IAA content.The release of dormancy of Notopterygium incisum seeds must meet the conditions of alternating warm and cold-low temperature changes.During the stratification process,the content of nutrients stored inside the seeds decreases,enzyme activity changed significantly,ABA content decreased,and GA 3 and IAA contents increased,which worked together to promote the release of dormancy of Notopterygium incisum seeds.
