摘要:目的 建立RP-HPLC法同时定量测定甘肃和云南当归药材中5种化学成分(阿魏酸、正丁基苯酞、正丁烯基苯酞、Z-藁本内酯、亚油酸),综合评价当归药材质量.方法 选用Waters X-Bridge C18(250 mm×4.6 mm,5μm)色谱柱,以甲醇-水溶液为流动相,梯度洗脱,体积流量为0.8 mL/min,检测波长为280 nm,柱温30℃,进样体积为10βL.采用主成分分析进行质量评价.结果 56批当归药材中5种化学成分在各自线性范围内呈良好的线性关系(r >0.999 8),方法平均回收率(n=9)在95.6% ~ 104.3%之间.结论 本法的色谱条件能够将5种成分完全分离并同时定量测定.主成分分析显示“岷归”质量最佳与道地药材结论一致,云南产当归(“云归”)质量亦佳.
AIM To establish a quantitative method for simultaneously determining five constituents in Angelica sinensis (ferulic acid, Z-ligustilide, 3-butylphthalide, n-butylidenephthalide and linolic acid) and make comprehen- sive quality evaluation of A. sinensis collected from Gansu and Yunnan Province, China. METHODS The separa- tion was performed on a column of Waters X-Bridge Cls (250 mm ×4.6 mm,5μm) by RP-HPLC. The mobile phase was methanol-water under a flow rate of 0. 8 mL/min in a gradient elution manner. The detection wavelength was set at 280 nm. The column temperature was 30 ℃. The injection volume was 10 μL. Principal component analy- sis (PCA) was used for quality evaluation. RESULTS The five constituents showed good linear correlations (r 〉 0. 999 8 ) and the analyzed samples were from fifty-six batches. The average recoveries were between 95.6% - 104. 3% (n = 9). CONCLUSION The established chromatography can completely separate five index constituents and simultaneous determine them. PCA shows that the A. sinensis from Min County, Gansu Province has the best quality in consistent with that of famous-region herb, and A. sinensis from Yunnan Province comes the second.
