摘要:目的探讨当归黄芪超滤膜提取物预处理对培养乳鼠心肌细胞缺氧/复氧损伤和氧化损伤的保护作用。方法体外培养原代乳鼠心肌细胞,不同分子量及剂量当归黄芪超滤膜提取物预处理24h后,采用连二亚硫酸钠(Na2S2O4)诱导心肌细胞缺氧/复氧损伤模型,测定细胞存活率(MTT法)、细胞培养液中乳酸脱氢酶(LDH)释放量、细胞内超氧化物歧化酶(SOD)活性和丙二醛(MDA)含量。结果在缺氧/复氧损伤模型和氧化损伤模型中,当归红芪超滤膜提取物预处理明显提高细胞存活率,增加胞内SOD活性,降低培养液中LDH释放量和胞内MDA含量。结论当归红芪超滤膜提取物预处理对缺氧/复氧损伤以及氧化损伤心肌细胞都具有明显的保护作用,其可能与防止氧自由基的产生或抑制氧自由基对心肌细胞的损伤有关。
Objective To observe the effects of preconditioning of the mixture of Angelica sinensis and Hedysarum on polybotrys cardiac myocyte injury by hypoxia/reoxygenation in neonatal rats. Methods The neonatal rats' cardiac myocytes were isolaled and cultured in vitro. After 24 hours' preconditioning by the mixture of Angelica sinensis and Hedysarum polybotrys, the model of hypoxia/reoxygenation injury was induced by sodium dithionite( Na2 S2O4 ). In the model rats, the viability of cardiac myocytes (using MTT method), dehydrogenase(LDH) activities in the medium, superoxide dismutase(SOD) activities and intracellular malonaldehyde (MDA) levels were measured. Results In the model rats of hypoxia/reoxygenation injury induced by Na2 S2 O4 , preconditioning of icariin could significantly raise the viability of cardiomyocytes and SOD activities, and decrease LDH leakage and intracellular MDA production. Conclusion Preconditioning of the mixture of Angelica sinensis and Hedysarum polybotrys can obviously protect cultured neonatal cardiac myocytes from hypoxia/reoxygenation injury, and the beneficial effects may be related to its antioxidant property.
