摘要:目的:探讨黄芪多糖对炎性微环境中骨髓间充质干细胞(BMSCs)增殖、分化及肿瘤相关成纤维细胞(TAFs)相关分子表达的影响。方法:采用IL-6和TNF-α模拟BMSCs体外炎性微环境培养体系,并以黄芪多糖最佳药物浓度干预该体系;通过CCK-8法、流式细胞术及Western Blot分别检测黄芪多糖对炎性微环境中BMSCs细胞增殖、周期及TAFs标记分子α-SMA、FAP蛋白的表达变化。结果:与正常对照组比较,炎性因子干预组细胞生长速度加快,G1期比例降低,S期比例升高,且α-SMA、FAP蛋白表达明显增加,均有显著性差异(P<0.05);与炎性因子干预组比较,黄芪多糖组(50mg/L)生长速度减慢,G1期比例升高,S期比例降低,且α-SMA、FAP蛋白表达有所降低(P<0.05)。结论:IL-6和TNF-α模拟炎性微环境中BMSCs形态、增殖特性发生异常改变,具有向肿瘤相关成纤维细胞分化性,且黄芪多糖可抑制炎性微微环境中BMSCs异常改变。
Objective: To study the effects of astragalus polysaccharide on bone marrow mesenchymal stem cells(BMSCs) proliferation, differentiation and tumor associated fibroblasts molecular expression under inflammatory microenvironment. Methods: Interleukin 6(IL-6) and tumor necrosis factor-α(TNF-α) were used to simulate BMSCs in vitro inflammatory microenvironment, and optimal concentration of astragalus polysaccharide was used for intervention of this system. Effects of astragalus polysaccharide on BMSCs proliferation, cycle as well as protein expression of biomarkers of TAFs, α-SMA and FAP were detected respectively by CCK-8 method, flow cytometry and Western Blot. Results: Compared with normal control group, inflammatory factor intervention group showed accelerated cell growth speed, reduced G1 phase and increased S phase proportion, and α-SMA, FAP protein expression significantly increased(P〈0.05). Compared with inflammatory factor intervention group, astragalus polysaccharide group(50 mg/L) showed slow growth speed, increased G1 phase and reduced S phase proportion, and α-SMA, FAP protein expression decreased(P〈0.05). Conclusion: There are abnormal changes of BMSCs morphology and proliferation characteristics, which was likely to differentiate into tumor related fibroblasts, and astragalus polysaccharide can inhibit abnormal changes of BMSCs in the inflammatory environment.
