摘要:糖尿病视网膜病变(DR)是常见的糖尿病微血管并发症,尚无根治手段。铁死亡是一种依赖铁及脂质过氧化的新型细胞死亡方式,在视网膜神经血管单元中作用突出。研究表明,铁死亡通过影响视网膜内皮细胞、色素上皮细胞、周细胞、神经元和胶质细胞的存活与功能,参与高糖和年龄相关的视网膜损伤,并与血-视网膜屏障破坏及神经退行性病变密切相关。本文综述铁死亡的分子机制及其对DR的视网膜神经血管单元组成细胞的调控作用,为DR的早期诊断和干预提供理论依据。
Diabetic retinopathy(DR)is a common microvascular complication of diabetes,for which no definitive cure is currently available.Ferroptosis,a novel form of cell death characterized by iron dependency and lipid peroxidation,plays a prominent role in the retinal neurovascular unit.Emerging evidence indicates that ferroptosis affects the survival and function of retinal endothelial cells,retinal pigment epithelial cells,pericytes,neurons,and glial cells,thereby contributing to hyperglycemia-and aging-related retinal injury.Moreover,ferroptosis is closely associated with disruption of the blood-retinal barrier and retinal neurodegeneration.This review summarizes the molecular mechanisms of ferroptosis and its regulatory effects on the constituent cells of the retinal neurovascular unit in DR,providing a theoretical basis for the early diagnosis and intervention of DR.
