摘要：目的观察不同厂家二甲基亚砜(DMSO)对体外培养的人肺腺癌A549细胞的最小毒性体积分数,为体外筛选抗肺肿瘤药物实验溶媒的选取提供依据。方法将DMSO以不同体积稀释到DMEM完全培养液中,孵育细胞后,显微镜观察细胞形态,采用四甲基偶氮唑蓝显色(MTT)法和流式细胞仪技术(FCM)法分别检测细胞相对活力、细胞凋亡及细胞周期。结果 Sigma公司DMSO体积分数≤0.1%时,肺腺癌A549细胞能够正常生长并保持形态完整,细胞活力与对照组比较差异无统计意义(P>0.05),细胞未发生凋亡;当体积分数达到4.0%时,细胞生长异常,细胞相对活力显著降低(P<0.01),细胞凋亡率梯度性增高,G1+S期细胞比例增加。国产DMSO对A549细胞的毒性较进口试剂弱,体积分数≤0.5%时,对A549细胞影响较小(P>0.05)。结论大剂量DMSO对A549细胞生长毒性明显,可抑制细胞增殖、诱导细胞凋亡以及影响细胞周期;Sigma公司、天津科密欧公司产品DMSO体积分数分别低于0.1%,0.5%时可作为人肺腺癌A549细胞体外筛选抗肿瘤药实验的安全溶媒。
Objective To observe the minimal toxic volume fractions of dimethyl sulfoxide( DMSO) from different manufacturers to lung adenocarcinoma A549 cells screened in vitro culture and then to provide basis for selecting the experimental solvent of anti-lung cancer drugs. Methods Different volumes of DMSO were diluted into the complete culture solution of DMEM. After hatching for 48 hours,cellular morphology was examined through microscope; the viability,apoptosis rate and cycle of cell lines were detected by MTT and FCM. Results When the volume fraction of DMSO from Sigma Reagent Co. was lower than 0. 1 %,A549 cells were able to grow normally and kept morphological integrity. Compared with the control group,there was no any difference in the viability of cells( P>0.05); no apoptosis was detected. When the volume fraction of DMSO was at 4.0%,cell growth was abnormal,and the viability decreased significantly( P < 0.01); the rate of apoptosis gradient increased and the proportion of cells in G1+ S phase increased. The toxicity of DMSO produced by domestic company on cancer cell lines was weaker than that of imported product; when the volume fraction was less than 0.5%,there was little effect on A549 cells( P>0.05). Conclusion The toxicity of DMSO in large doses is noticeable and it could inhibit cell proliferation,induce apoptosis and affect cell cycle. When the volume fractions of DMSO were lower than 0.1%( Sigma Reagent Co.) and 0.5%( Tianjin Kemiou Chemical Reagent Co.) respectively,the reagents can be used as safe solvent for screening the lung adenocarcinoma A549 cells in the experiment of anti-lung cancer drugs.
