摘要：目的研究当归补血汤(DBD)对溃疡性结肠炎大鼠的治疗作用及其对肠黏膜屏障的修复作用。方法 Wistar雄性大鼠按体重随机分为6组,每组8只。用三硝基苯磺酸制备溃疡性结肠炎大鼠模型。正常组与模型组动物用生理盐水灌胃,对照组大鼠灌胃给予柳氮磺吡啶(SASP 0.5 g·kg^(-1)),3个剂量DBD实验组分别灌胃DBD 3.6,7.2和14.4 g·kg^(-1),连续给药14 d。考察大鼠病变活动指数(DAI)变化;收集结肠标本进行病理组织学评分,杯状细胞计数及髓过氧化物酶(MPO)活性测定;酶联免疫吸附法测定血清D-乳酸(D-LA)及二胺氧化酶(DAO)含量。结果与正常相比,模型组大鼠DAI、病理学评分、MPO活性、杯状细胞数量及血清D-LA和DAO水平均明显增高(P<0.05)。高、中2个剂量DBD组可显著降低模型大鼠DAI、病理学评分、MPO活性、杯状细胞数量及血清D-LA和DAO水平(P<0.05)。结论 DBD可改善溃疡性结肠炎大鼠肠道上皮细胞黏膜屏障功能,对溃疡性结肠炎具有治疗作用。
Objective To study the effect of Danggui Buxue decoction（ DBD） on intestinal mucosal barrier function in ulcerative colitis rats.Methods Wistar male rats were randomly divided into 6 groups,ulcerative colitis rat model was established by colonic 2,4,6-trinitrobenzenesulfonic acid sol（ TNBS） administration.Rats in normal and model groups were orally administered with saline,rats in control group were orally treated with salazosulfapyridine（ SASP） at the dose of 0.5 g · kg^-1,while rats in DBD groups were orally administered with DBD at the dose of 3.6,7.2 and 14.4 g·kg^-1,respectively.All the drugs were administered to rats for consecutive 14 d.Disease activity index（ DAI） was investigated during the drug treatment,colon sample were collected after drug administration for evaluating histopathological score,myeloperoxidase（ MPO） activity and goblet cell number,and blood sample were also collected for determination of serum level of D-lactic acid（ D-LA）and diamine oxidase（ DAO） by ELISA.Results Compared with normal group,DAI value,histopathological score,MPO activity,goblet cell number as well as serum D-LA and DAO level in model group significantly increased（ P〈0.05）.Compared with the model group,large andmedium dose of DBD group significantly reduced the DAI value,histopathological score,MPO activity,goblet cell number as well as serum D-LA and DAO level in colitis rats（ P〈0.05）.Conclusion DBD treatment has effect on ulcerative colitis rats via improving the function of intestinal mucosal barrier.
