摘要:目的建立党参休眠芽玻璃化超低温保存技术体系。方法采用单因子试验设计对党参休眠芽的预培养、PVS2处理时间及苞片有无、低温锻炼时间等几个关键因素进行研究,以建立党参休眠芽玻璃化超低温保存体系。结果将两年生的新鲜党参覆土后置于4℃低温锻炼90d左右,切取带少量根基和苞片的完整休眠芽,先用2mol/L甘油+0.4mol/L蔗糖溶液装载20min,再用PVS2处理80min后直接投入液氮。恢复培养时从液氮中取出,先用40℃水浴化冻2min,再用含1.2mol/L蔗糖的MS无机盐培养液卸载20min,最后用无菌滤纸吸干芽表面的溶液,接种到恢复生长培养基中(MS+6-BA 1.0mg/L+NAA 0.5mg/L),于正常光照下培养,休眠芽冻存后的成活率达到74%。结论建立了党参休眠芽玻璃化超低温保存技术体系,该技术方法可为药用植物种质资源保存开辟一条新途径。
Objective The study aimed at investigating the cryopreservation of Codonopsis pilosula(Franch.)Nannf. dormant buds by vitrification. Methods Dormant buds as an experimental material, the influence of preculture, PVS2 processing times, bract, cold exercising time and etc. on cryopreservation of Codonopsis pilosula(Franch.)Nannf. were studied.Results The optimal procedures were as follows:Biennials fresh Codonopsis pilosula(Franch.)Nannf. were exercised with covering soil about 90 d, and dormant buds were cut with little root and bract. The buds were immersed in loading solution for 20 min at 20℃, and then treated in PVS2 solution for 80 min at 0℃ and finally plunged into liquid nitrogen quickly. After 24 hours, the buds were rapidly thawed in a water bath at 40℃ for 2 min and washed twice with 1/2 MS liquid medium containing 1.2 mol/L sucrose. Finally the buds were transferred to regeneration medium(MS+1.0 mg/L 6-BA+0.5 mg/L NAA), the survival rate was up to 74%.Conclusion A high-efficiency cryopreservation protocol of Codonopsis pilosula(Franch.)Nannf. was set up, which could provide a new way for preservation for germplasm of medicinal plants.
