摘要:目的观察艾灸对菌群失调幼鼠结肠黏膜屏障的影响,探讨艾灸对结肠黏膜的保护作用及机制。方法50只SPF级雄性Balb/c幼鼠按随机数字表法分为空白组、模型组、阳性药组、艾灸组和联合组,每组10只。除空白组外,其余各组采用混合抗生素灌胃诱导菌群失调。造模14 d后干预,模型组每日固定束缚15 min;阳性药组予双歧杆菌四联活菌片0.68 g/kg灌胃;艾灸组艾灸“神阙”“中脘”“关元”“天枢”,15 min/次,1次/d;联合组在艾灸同时予双歧杆菌四联活菌片。连续干预14 d后,采集幼鼠粪便,采用16S rDNA测序检测肠道菌群多样性,腹腔注射脂多糖诱导肠道炎症反应,HE染色观察幼鼠结肠黏膜完整性,Western blot检测结肠组织Occludin、ZO-1蛋白表达。结果与空白组比较,模型组幼鼠肠道菌群丰富度指数Chao1、ACE及多样性指数Shannon显著降低(P<0.01),多样性指数Simpson显著升高(P<0.01);与模型组比较,艾灸组、阳性药组和联合组幼鼠肠道菌群多样性指数Shannon显著升高(P<0.01),多样性指数Simpson显著降低(P<0.01)。HE染色显示,与空白组比较,模型组幼鼠结肠黏膜损伤严重;与模型组比较,各治疗组幼鼠结肠黏膜损伤减轻,联合组幼鼠结肠黏膜损伤减轻最明显。Western blot结果显示,与空白组比较,模型组幼鼠结肠组织Occludin、ZO-1蛋白表达显著降低(P<0.01);与模型组比较,艾灸组、阳性药组和联合组幼鼠结肠组织Occludin、ZO-1蛋白表达显著升高(P<0.05,P<0.01);与艾灸组及阳性药组比较,联合组幼鼠结肠组织Occludin、ZO-1蛋白表达显著升高(P<0.01)。结论艾灸可保护菌群失调幼鼠结肠黏膜,其机制可能与增加肠道菌群多样性、维持肠道菌群结构稳定及保护结肠黏膜屏障有关。
Objective To observe the effects of moxibustion on intestinal flora imbalance in juvenile mice;To explore the protective effect and mechanism of moxibustion on colonic mucosa.Methods Totally 50 male SPF Balb/c juvenile mice were randomly divided into blank group,model group,positive drug group,moxibustion group and combined group,with 10 rats in each group.Except the blank group,the other groups were given mixed antibiotics to induce dysbacteriosis.14 days after modeling,the model group was fixed and restrained for 15 minutes every day;the positive drug group was given Bifidobacterium quadruple viable bacteria tablet 0.68 g/kg for gavage;the moxibustion group received moxibustion in“Shenque”,“Zhongwan”,“Guanyuan”and“Tianshu”,15 min/time,1 time/d;the combined group was given Bifidobacterium quadruple viable tablet while moxibustion.After the intervention for 14 consecutive days,the feces of juvenile mice were collected,16S rDNA sequencing was used to detect the diversity of intestinal flora,and intraperitoneal injection of lipopolysaccharide was for inducing intestinal inflammation,HE staining was used to observe the integrity of the colonic mucosa,Western blot was used to detect the expressions of Occludin and ZO-1 protein in colon tissue.Results Compared with the blank group,the intestinal flora richness index Chao1,ACE and diversity index Shannon of the juvenile mice in model group significantly decreased(P<0.01),and the diversity index Simpson significantly increased(P<0.01).Compared with the model group,the diversity index Shannon in the intestinal flora of the juvenile mice in moxibustion group,positive drug group and the combined group significantly increased(P<0.01),and the diversity index Simpson significantly decreased(P<0.01).HE staining showed that compared with the blank group,the colonic mucosa in the model group was seriously damaged.Compared with the model group,the colonic mucosal damage of the juvenile mice in each treatment group was alleviated,and the colonic mucosa damage of the juvenile mice in the combined group was most significantly alleviated.Western blot results showed that compared with the blank group,the expressions of Occludin and ZO-1 protein in colon tissue of the juvenile mice in model group decreased significantly(P<0.01).Compared with the model group,the expressions of Occludin and ZO-1 protein in colon tissue of the juvenile mice in moxibustion group,positive drug group and combined group significantly increased(P<0.05,P<0.01).Compared with the moxibustion group and positive drug group,the expressions of Occludin and ZO-1 protein in the combined group increased significantly(P<0.01).Conclusion Moxibustion can protect colonic mucosa of intestinal flora imbalance in juvenile mice,and its mechanism may be related to increasing the diversity of intestinal flora,maintaining the stability of flora structure and strengthening the barrier of colonic mucosa.
