摘要:本文通过研究黄芪主要成分对转化生长因子-β1(TGF-β1)诱导下睾丸间质细胞(Leydig cell)功能和形态变化的影响,探讨黄芪主要成分中对睾丸Leydig细胞增殖和活性影响具有最佳作用的成分。采用体外培养睾丸Leydig细胞,将培养的Leydig细胞分为3组:模型组(5 ng·mL^(-1)TGF-β1造模)、中药干预组、正常对照组。中药干预组分别加入终浓度10μg·mL^(-1)、50μg·mL^(-1)黄芪总多糖,终浓度10μg·mL^(-1)、50μg·mL^(-1)黄芪总黄酮,终浓度20μg·mL^(-1)、100μg·mL^(-1)黄芪总皂苷。ELISA法检测T、E2蛋白含量和细胞爬片。使用SPSS20.0软件对数据进行统计分析和显微镜观察细胞爬片。结果表明,TGF-β1干预使Leydig细胞形态异常,数量减少,明显抑制Leydig细胞合成T、E2,与正常组比较差异显著(P≤0.05)。黄芪总多糖、黄芪总黄酮、黄芪总皂苷都均能改善睾丸Leydig细胞病变,促进Leydig细胞的增殖,提高TGF-β1损伤的睾丸Leydig细胞的活性,与模型组比较差异显著(P≤0.05)。终浓度100μg·mL^(-1)黄芪总皂苷促进TGF-β1诱导的睾丸Leydig细胞增殖作用最明显,终浓度20μg·mL^(-1)的黄芪总皂苷缓解TGF-β1对睾丸Leydig细胞合成T、E2的抑制作用最明显。结论:黄芪总皂苷对TGF-β1诱导的睾丸Leydig细胞增殖和活性影响效果最显著。
By studying the effects of different astragalus extracts on functions and morphological changes of TGF-β1-induced leydig cells in testis,the research is designed to find out the very composition of the extract that dominates proliferation and activity of TGF-β1-induced leydig cells in testis.To divide the leydig cells cultured in vitro into three groups:model group(5 ng·mL^(-1)TGF-β1 modeling),Chinese medicine intervention group and normal control group.the final concentration of 10μg·mL^(-1)and 50μg·mL^(-1)total Astragalus polysaccharide,final concentration of 10μg·mL^(-1)and 50μg·mL^(-1)total astragalus flavonoids,final concentration of 20μg·mL^(-1)and 100μg·mL^(-1)total astragalus saponins were added to the intervention group of traditional chinese medicine.ELISA detection of T、E2 protein content and cell climbing tablets.The results showed that TGF-β1 intervened Leydig cells were abnormal in morphology,decreased in number inhibiting T and E2 composition in Leydig cells,producing a significant difference(P≤0.05)compared with the normal group.Total polysaccharides,astragalys flavonoids,astragalus saponins all proved to be effective in lessening lesions and promoting proliferation in Leydig cells in testis,the larger the amount,the better the effects.Furthermore,the three additives all improved activity of TGF-β1-induced Leydig cells in testis with a significant difference(P≤0.05)compared with the model group.The final concentration of 100μg·mL^(-1)astragalus total saponins promoted TGF-β1 induced testicular Leydig cell proliferation,and the final concentration of 20μg·mL^(-1)of astragalus total saponins alleviated TGF-β1 the inhibition of testicular Leydig cell synthesis was the most obvious.Total astragaloside has the most significant effect on TGF-β1 induced testicular Leydig cell proliferation and activity.
