摘要:目的研究平痫冲剂对幼龄癫痫大鼠海马及皮质区神经元ICAM-1、Cx43蛋白表达的影响,探讨其抗痫的相关作用机制。方法 28天龄Wistar大鼠在癫痫模型建成后随机分为空白组、模型组、西药组(0.36g/kg)、平痫冲剂组(3.22g/kg),每组60只。连续灌胃3周后取材,HE染色观察大鼠海马神经元的形态变化,免疫组织化学法观察大鼠海马神经元ICAM-1、Cx43蛋白表达的变化。结果治疗21天后,HE染色显示西药组、平痫冲剂组与模型组比较,神经元细胞水肿消退,核形态恢复完整,细胞形态恢复趋向正常组织。免疫组化显示西药组、平痫冲剂组与模型组比较ICAM-1、Cx43阳性表达减弱(P<0.01);另外西药组、平痫冲剂组在用药24h后与模型组相比较,皮质区ICAM-1、Cx43的MOD值即下降(P<0.05);至用药72h始,与上一时间点相比较,皮质区ICAM-1、Cx43的MOD值明显下降(P<0.01);与西药组比较,平痫冲剂组皮质区MOD值无统计学意义(P>0.05)。结论平痫冲剂可能是通过下调ICAM-1、Cx43蛋白表达进而影响BBB的结构与功能发挥抗痫作用。
Objective To investigate the effects of Pingxian Granule on the expression of ICAM-1 and Cx43 in hippocampus and cortex of young rats with epilepsy,and to explore the related mechanism of anti-epilepsy.Methods After the epilepsy model was established,28-day-old Wistar rats were randomly divided into the blank group,the model group,the western medicine group(0.36 g/kg),and the Pingxian granule group(3.22 g/kg),with 60 rats in each group.After 3 weeks of continuous gavage,the morphological changes of hippocampal neurons were observed by HE staining.The expressions of ICAM-1 and Cx43 in hippocampal neurons were observed by immunohistochemistry.Results After 21 days of treatment,HE staining showed that compared with the model group,the neuronal cell edema subsided,the nuclear morphology recovered,and the cell morphology recovered to normal tissues.Immunohistochemistry showed that the positive expressions of ICAM-1 and Cx43 were decreased in the Western medicine group and Pingxian granule group compared with the model group(P<0.01).In addition,the western medicine group and Pingxian granule group were compared with the model group after 24 hours of treatment,and the cortical area ICAM was compared.The MOD value of ICAM-1 and Cx43 decreased(P<0.05).Compared with the previous time point,the MOD values of ICAM-1 and Cx43 in the cortical area decreased significantly(P<0.01).Compared with the western medicine group.There was no significant difference in MOD between the corticosteroids of Pingxian Granules group(P>0.05).Conclusion Pingxian granules may exert anti-epileptic effects by down-regulating the expression of ICAM-1 and Cx43 proteins and affecting the structure and function of BBB.
