摘要:目的研究四逆散干预创伤后应激障碍(PTSD)大鼠海马组织的分子生物学机制。方法按照随机数表法将SD大鼠分为5组:空白对照组、模型组、阴性对照组、阳性对照组、实验组,每组10只。除空白对照组外,其他4组动物均复制PTSD模型。空白对照组不接受药物干预,模型组不接受药物治疗,阴性对照组灌胃等体积0. 9%Na Cl,阳性对照组灌胃盐酸帕罗西汀溶液0. 42mg·m L^(-1),实验组灌胃四逆散水煎液(含生药0. 24 g·m L^(-1)),于应激造模前1h灌胃给药10 m L·kg^(-1),均1天/次,共计7 d。于末次给药5 h后,采集海马组织。用代谢组学方法筛选大鼠海马组织特征性分子。结果模型组与空白对照组比较,海马组织共存在14个差异离子:维生素A、硫化孕烯醇酮、磷酸烯醇丙酮酸、磷脂酰甘油、棕榈酸、苯乙胺、2,6-二甲基苯胺、鞘氨醇、神经鞘氨醇、链甾醇、酵母甾醇、异戊烯基膦酸、7-脱氢胆固醇和芥子酸,除芥子酸含量降低外,其余的含量均升高。实验组与模型组比较,共存在10个差异分子:鞘氨醇、神经鞘氨醇、芥子酸、磷酸烯醇丙酮酸、硫化孕烯醇酮、异戊烯基膦酸、亚油酸、7-脱氢胆固醇、链甾醇和酵母甾醇,除芥子酸含量升高外,其余的含量均下降。结论 PTSD大鼠海马组织差异分子存在不同程度的上调或下调,这些代谢分子的异常表达可能是PTSD的重要神经内分泌机制。
Objective To study the function mechanism of the Sinisan on hippocampus molecular expression in rats with post-traumatic stress disorder( PTSD). Methods Sprague-Dawley rats were divided equally into five groups: blank control group,model group,negative control group,positive control group and experimental group. The control blank group did not copy the model,did not receive treatment but normal feeding. Model group,repetitive PTSD model was induced by current stimulation,but not treated. Negative control group,equal volume of 0. 9%NaCl. Positive control group( paroxetine hydrochloride 0. 42 mg·m L-1) and experimental group( Sinisan,containing crude drug 0. 24 mg·m L-1). It given to drugs( 10 m L·kg-1) 1 h before the model establishment. Once a day,for a total of 7 d. At the end of the last administration for 5 h,hippocampal tissue was collected. Screening of characteristic molecules in rat hippocampus by metabonomics. Results Compared with blank control group,there were fourteen materials were identified by database,and concentration increased. Except for the decrease of erucic acid content,the others increased. That were vitamin A,pregnenolone sulfide,phosphoenolpyruvate,phosphatidyl glycerol,palmitic acid,phenylethylamine,2,6-dimethylaniline,sphingosine,nerve sphingosine,desmosterol,zymosterol,isopentenylphosphonic acid,7-dehydrocholesterol,and erucic acid. Compared with model group,there was no significant difference in the negative control group. Compared with negative control group,there were significant differences between the two drug groups. There were ten differential metabolites in the experimental group,That were,sphingosine,neurilinol,erucic acid,phosphoenolpyruvate,pregnenolone sulfide,isopentenyl phosphonic acid,linoleic acid 7-dehydrocholesterol,streptosterol,yeast sterol,and erucic acid,except for the increase of erucic acid content,the rest all decreased.Conclusion Differential molecules in hippocampus of PTSD rats were up-regulated or down-regulated in varying degree,abnormal expression of these metabolic molecules may be an important neuroendocrine mechanism of PTSD.
