摘要:目的研究藏药黑果枸杞对电刺激蟾蜍离体腓肠肌收缩功能的影响。方法取20条蟾蜍腓肠肌,随机将其分为4组:对照组和高、中、低3个剂量实验组,每组5条。对照组腓肠肌浸泡在25 mL任氏液中,实验组腓肠肌分别浸泡在终浓度为1. 2,0. 6,0. 3 g·mL^(-1)的黑果枸杞水煎液25 mL中,15 min后取出腓肠肌。用脉冲式电流刺激腓肠肌,以生物机能分析系统记录收缩幅度达最大收缩幅度50%和10%所需要的时间(T50%和T10%),用无钙任氏液替代普通任氏液,并加入钙通道阻断药氯化铬重复实验。结果对照组和低、中、高3个剂量实验组腓肠肌的阈刺激强度分别为(0. 30±0. 02),(0. 35±0. 04),(0. 40±0. 03),(0. 46±0. 08) V;这4组的腓肠肌的饱和刺激强度分别为(1. 00±0. 05),(1. 10±0. 08),(1. 20±0. 04),(1. 28±0. 02) V。腓肠肌达最大肌肉收缩幅度所需时间:这4组分别为(235. 00±3. 47),(218. 00±5. 25),(205. 00±6. 24),(176. 00±3. 87) s。这4组的T50%分别为(125. 00±3. 45),(134. 00±3. 42),(254. 00±2. 59),(304. 00±3. 79) s;这4组的T10%分别为(268. 00±7. 09),(380. 00±6. 14),(521. 00±7. 74),(552. 00±7. 73) s。3个剂量实验组与对照组比较,以上全部指标差异均有统计学意义(均P <0. 05)。加入钙通道阻断药氯化铬后,该缓解作用明显降低。结论藏药黑果枸杞可增强蟾蜍离体腓肠肌的收缩能力,对电刺激引发的收缩疲劳有缓解作用,其作用机制可能与激动"钙库",促进钙离子内流,增加细胞内钙离子浓度有关。
Objective To investigate the effect of Tibetan Lycium ruthenicum Murr( LM) decoction on the contractility of isolated gastrocnemius muscle stimulated by electric stimulation. Methods Twenty gastrocnemius specimens were randomly divided into 4 groups: Control group,and high-dose,middle dose,and low-dose experimental groups,5 specimens in each group. The gastrocnemius in the control group was immersed in 25 mL Ren’s solution,and the gastrocnemius in the experimental groups was immersed in 25 mL LM decoction with final concentration of 1. 2,0. 6,0. 3 g mL-1 respectively. After 15 min,the gastrocnemius was removed and stimulated by pulsed current. The time needed to reach the maximum contraction rate of 50% and 10% was recorded( T50%and T10%). The mechanism was discussed by adding calcium free medium and calcium channel blocker. Results The threshold stimulation intensity of gastrocnemius in control group,low-dose,middle dose,and high-dose experimental groups were( 0. 30 ± 0. 02),( 0. 35 ± 0. 04),( 0. 40 ± 0. 03),( 0. 46 ± 0. 08) V respectively;the saturation stimulation intensity of gastrocnemius in the 4 groups were( 1. 00 ± 0. 05),( 1. 10 ± 0. 08),( 1. 20 ± 0. 04),( 1. 28 ± 0. 02) V respectively. The time required for maximum muscle contraction in the 4 groups were( 235. 00 ± 3. 47),( 218. 00 ± 5. 25),( 205. 00 ± 6. 24),( 176. 00 ± 3. 87) s,respectively. T50%in the 4 groups were( 125. 00 ± 3. 45),( 134. 00 ± 3. 42),( 254. 00 ± 2. 59),( 304. 00 ± 3. 79) s,respectively;T10%in the 4 groups were( 268. 00 ± 7. 09),( 380. 00 ± 6. 14),( 521. 00 ± 7. 74),( 552. 00 ± 7. 73) s,respectively. Comparing between three doses experimental groups with the control group,the difference of the all factors were significant( all P < 0. 05). After calcium channel blockers added,the mitigative effect decreased obviously. Conclusion Lycium ruthenicum Murr decoction could enhance the contractility of toad gastrocnemius in vitro,and alleviate the contraction fatigue induced by electrical stimulation,which might be related to the promotion of calcium influx and the increase of intracellular calcium concentration.
