摘要:目的探讨^(60)Coγ射线对性成熟期大鼠外周血细胞和性激素水平以及卵巢组织凋亡的影响以及左归丸的干预效应。方法选择10~12周龄雌性SPF级SD大鼠50只,随机分成正常组、辐射组、补佳乐组、左归丸高剂量组和左归丸低剂量组。^(60)Coγ射线一次性照射(6.0 Gy,LD40)24 h后连续给补佳乐[0.09 g/(kg·d)]、左归丸高剂量(0.946 g生药/200 g)和左归丸低剂量(0.189 g生药/200 g),1次/d,治疗21 d,检测不同时间段血细胞变化、EILSA法检测外周血激素[促卵泡生成素(follicle-stimulating hormone,FSH)、黄体生成素(luteinizing hormone,LH)、雌二醇(estradiol,E_(2))],光镜下过程卵巢的组织形态变化,用免疫组化检测卵巢组织蛋白TAP63和Bcl-2蛋白表达。结果辐射后大鼠外周血细胞数量减少,尤以白细胞减少最显著,辐射组与对照组比较有显著统计学意义(P<0.01),辐射照组成熟卵泡数量减少,卵巢血管增生,有纤维化,E_(2)[(49.40±12.54)pg/mL]降低,FSH[(1.97(0.78))pg/mL]和LH[(1.65±1.57)pg/mL]水平增加,之后FSH[(0.55±0.49)pg/mL]和LH[(0.704±0.093)pg/mL]逐渐降低(P<0.01);TAP63和Bcl-2蛋白表达明显减(P<0.01),在给予补佳乐和左归丸治疗后逐渐恢复正常,尤以左归丸高剂量组作用显(P<0.01)。结论辐射损伤抑制骨髓造血、促进卵巢颗粒细胞凋亡和卵泡闭锁,导致卵巢功能快速衰退,左归丸通过刺激骨髓造血,促进卵巢组织TAP63蛋白表达,激活Bcl-2基因抑制颗粒细胞和卵母凋亡,增加颗粒细胞合成雌激素,达到促进卵巢功能修复的作用。
Objective To investigate the effects of^(60)Coγon the levels of peripheral blood cells,sex hormones and ovarian tissue apoptosis in sexually mature rats and the intervention effect of Zuogui Pill(左归丸).Methods Fifty SPF female SD rats aged 10-12 weeks were randomly divided into the normal group,the radiation group,the Progynova group,Zuogui Pill high-dose group and Zuogui Pill low-dose group.After^(60)Coγone-time exposure(6.0 Gy,LD40)for 24 h,Progynova[0.09 g/(kg·d)],Zuogui Pill high dose(0.946 g/200 g)and low dose(0.189 g/200 g)were given once a day,continuously for 21 d.The blood cell analysis was tested.EILSA method was used to detect peripheral blood hormones[follicle-stimulating hormone(FSH),luteinizing hormone(LH),estradiol(E_(2))]and the ovarian tissue morphological changes,TAP63 and Bcl-2 protein expressions were observed.Results After radiation,the peripheral blood cells were decreased,especially the white blood cells.The changes of the radiation group were obviously significantly compared with those of the control group(P<0.01).In the radiation group,the number of mature follicle was decreased and there was ovarian vascular proliferation and fibrosis.Th level of E_(2)[(49.40±12.54)pg/mL]was reduced and the levels of FSH[(1.97(0.78))pg/mL]and LH[(1.65±1.57)pg/mL]were increased.And then,the levels of FSH[(0.55±0.49)pg/mL]and LH[(0.704±0.093)pg/mL]were gradually reduced(P<0.01).The protein expressions of TAP63 and Bcl-2 were significantly decreased(P<0.01),and gradually recovered to be normal after treatment with Progynova and Zuogui Pill,especially in the Zuogui Pill high-dose group(P<0.01).Conclusion Radiation injury inhibits e tbone marrow hematopoiesis,promotes the ovarian granulosa cell apoptosis and follicular atresia,leading to the rapid decline of ovarian function.Zuogui Pill can stimulate the bone marrow hematopoiesis,promote the expressions of TAP63 protein in ovarian tissue,activate Bcl-2 gene,inhibit te granulosa cell and oocyte apoptosis,increase the synthesis of estrogen by granulosa cells and promote the repair of ovarian function.
