摘要:目的探讨不同分子量红芪多糖(HPS HG-2,HPS HG-3和HPS HG-4)对小鼠溃疡性结肠炎的保护作用机制。方法按照随机数字表法将小鼠随机分为7组:空白组、模型组、柳氮磺吡啶组、红芪总多糖(THPS)组、HPS HG-2,HPS HG-3和HPS HG-4组,每组10只。用2,4,6-三硝基苯磺酸灌肠来复制溃疡性结肠炎动物模型。造模后,空白组与模型组灌胃蒸馏水;柳氮磺吡啶组灌胃500 mg·kg^(-1)柳氮磺吡啶,THPS组灌胃THPS 147.2 mg·kg^(-1),HG-2、HG-3和HG-4组依次灌胃(HPS HG-2,HPS HG-3和HPS HG-4)29.5,33.2和36.0 mg·kg^(-1),1次/天,连续6 d。用苏木精-伊红染色对结肠组织病理学变化进行综合评分,用酶联免疫吸附法测定血清肿瘤坏死因子(TNF-α)、白细胞介素-6(IL-6)和IL-10的含量。结果空白组、模型组、柳氮磺吡啶组、THPS组、HPS HG-2,HPS HG-3和HPS HG-4组的结肠组织病理学变化综合评分分别为(0.12±0.18),(0.92±0.18),(0.39±0.18),(0.46±0.18),(0.54±0.18),(0.57±0.18)与(0.50±0.18)分;上述7组的TNF-α水平分别为(92.36±23.76),(446.36±60.44),(205.07±37.92),(229.07±41.00),(289.50±48.85),(319.29±51.31)与(276.43±45.39)pg·mL^(-1);上述7组的IL-6水平分别为(135.57±24.77),(423.36±67.38),(257.29±47.54),(267.14±46.18),(299.29±55.64),(325.64±59.90)与(291.57±50.36)pg·mL^(-1);上述7组的IL-10水平分别为(252.09±26.85),(131.82±16.53),(200.27±21.48),(215.27±25.79),(187.32±18.84),(173.95±20.05)与(199.86±23.07)pg·mL^(-1)。模型组的上述指标与空白组比较,差异均有统计学意义(均P<0.01);THPS、HPS HG-2,HPS HG-3和HPS HG-4组与模型组比较,差异均有统计学意义(P<0.05,P<0.01)。结论HPS HG-2,HPS HG-3和HPS HG-4对溃疡性结肠炎有治疗作用,缓解TNF-α、IL-6和IL-10异常变化是其作用机制之一。
Objective To investigate the protective effect of Hedysari Polysaccharides(HPS)and HPS with different molecular weights(HPS HG-2,HPS HG-3 and HPS HG-4)on ulcerative colitis mice and its mechanism.Methods Mice were randomly divided into 7 groups according to random number table:blank group,model group,sulfasalazine group(sulfasalazine 500 mg·kg^(-1)),total HPS(THPS)group(147.2 mg·kg^(-1)THPS),HPS HG-2 group(29.5 mg·kg^(-1)HPS HG-2),HPS HG-3 group(33.2 mg·kg^(-1)HPS HG-3)and HPS HG-4 group(36.0 mg·kg^(-1)HPS HG-4),10 mice in each group.Mice model of ulcerative colitis was replicated with 2,4,6-trinitrobenzenesulfonic acid.Mice in blank group and model group were orally administered with distilled water.All the mice were administered for consecutive 6 d,1 times a day.The pathological changes of colon were observed by hematoxylin eosin staining,and the comprehensive score was made.The level of tumor necrosis factorα(TNF-α),interleukin-6(IL-6)and interleukin-10(IL-10)in the serum were determined by enzyme linked immunosorbent assay.Results Comprehensive score of pathological changes in blank group,model group,sulfasalazine group,THPS group,HPS HG-2 group,HPS HG-3 group and HPS HG-4 group were(0.12±0.18),(0.92±0.18),(0.39±0.18),(0.46±0.18),(0.54±0.18),(0.57±0.18)and(0.50±0.18)point,respectively;the serum TNF-αin the above 7 groups were(92.36±23.76),(446.36±60.44),(205.07±37.92),(229.07±41.00),(289.50±48.85),(319.29±51.31),(276.43±45.39)pg·mL^(-1),respectively;the serum IL-6 in the above 7 groups were(135.57±24.77),(423.36±67.38),(257.29±47.54),(267.14±46.18),(299.29±55.64),(325.64±59.90)and(291.57±50.36)pg·mL^(-1),respectively;the serum IL-10 in the above 7 groups were(252.09±26.85),(131.82±16.53),(200.27±21.48),(215.27±25.79),(187.32±18.84),(173.95±20.05)and(199.86±23.07)pg·mL^(-1),respectively.Compared between model group and blank group,the difference of the factors were significant(all P<0.01);compared between THPS group,HPS HG-2 group,HPS HG-3 group,HPS HG-4 group and model group,the difference of the factors were significant(P<0.05,P<0.01).Conclusion THPS and HPS with different molecular weights(HG-2,HG-3 and HG-4)have therapeutic effects on ulcerative colitis.The effect of HPS in the treatment of ulcerative colitis is related to the alleviation of abnormal changes in TNF-α,IL-6 and IL-10.
