摘要:目的探讨当归对阴虚哮喘小鼠的平喘作用及对水通道蛋白(AQP)1、AQP5表达的影响。方法将小鼠随机分为正常对照组、模型对照组、地塞米松组、3个当归剂量组、当归与地塞米松组,通过注射卵清白蛋白(OVA)致敏、吸入OVA激发复制哮喘模型,实验后期给予甲状腺素复制成阴虚哮喘BALB/c小鼠模型,观测当归对哮喘发作鼠数、肺液清除功能、肺组织AQP1、AQP5及其基因表达的影响。结果 2、4、8 g/kg当归可明显减少哮喘发作鼠数,促进肺水代谢平衡,促进肺组织AQP1、AQP5及其基因的表达(P<0.05,0.01)。同时,当归与地塞米松配伍后对AQP1、AQP5及其基因表达有一定的协同作用(P<0.05)。结论当归治疗哮喘的机制之一是调节AQP1、AQP5及其基因表达而促进肺水代谢平衡。
Objective To study the effects of Radix Angelicae Sinensis (RASI) on anti-asthma and the expressions of aquaporin-1 (AQP1) and aquaporin-5 (AQP5) in lung tissue of asthmatic mice with Yin deficiency syndrome. Methods The BALB/c mice were randomly divided into control, model, Dexamethasone, three doses of RASI, middle dose of RASI and Dexamethasone groups. Asthmatic model mouse was made by injecting ovalbumin (OVA) to sensitize, then inhaling OVA to stimulate, and thyroxin was used to make asthmatic mouse with Yin deficiency syndrome during late stimulation. The number of animals with asthma attacks, pulmonary fluid clearance, and expressions of AQP1 and AQP5 and their genes in lung tissue were observed. Results 2,4 and 8 g/kg RASI could reduce the number of animals with asthma attacks,increase the ratio of W/D, and promote the expressions of AQP1 and AQP5 and their genes in lung tissue(P〈0. 05, P〈0. 01 ). In addition,these was a certain synergy between RASI and dexamethasone on improving the expressions of AQP1 and AQP5 and their genes (P〈 0. 05). Conclusions One of mechanisms of RASI/s anti-asthmatic effect is to regulate the AQP1 ,AQP5 proteins and their genes expression to promote water metabolism balance in lung.
