摘要:目的:研究黄芪百合颗粒对高原低氧模型小鼠肺自由基代谢及其病理学形态改变的影响,探讨黄芪百合颗粒预防高原反应的效应机制。方法:将SPF级小鼠采用随机数字表法分为空白对照组,模型组,黄芪百合颗粒低、中、高剂量组。灌胃给药,每天1次,连续14 d。除对照组外,第15天起各组小鼠每天灌胃30 min后于低氧舱中模拟高海拔环境进行低氧暴露72 h。末次出舱称重后处死小鼠。比色法检测各组小鼠过氧化脂质降解产物丙二醛(malondialdehyde,MDA)的含量及总抗氧化能力(total antioxidantcapacity,T-AOC)、过氧化物歧化酶(super-oxide dismutase,SOD)的活性。HE染色法观察小鼠肺组织形态学改变情况。结果:与空白对照组比较,模型组小鼠体质量下降;肺脏、肾脏及心脏指数均升高;肺MDA的含量明显升高,SOD及T-AOC的活性明显下降;小鼠肺病理评分结果和肺炎性细胞数量均明显升高(P<0.01)。与模型组比较,黄芪颗粒中、高剂量组小鼠有效体质量升高,肺脏、肾脏及心脏指数均降低;肺MDA含量下降,SOD及T-AOC活性均升高;小鼠肺病理评分结果和肺炎性细胞数量均下降(P<0.05)。结论:黄芪百合颗粒通过调节自由基代谢明显增强了其抗高原肺水肿的作用。
Objective:To study the effects of astragalus and lily particles on free radical metabolism and the lung pathological change in high altitude hypoxia mice,and to investigate its mechanism. Methods:SPF mice were randomized into control group,model group,low dose group of astragalus and lily granules,medium dose group of astragalus and lily granules,high dose group of astragalus and lily granules. They were intervened by intragastric infusion once a day for 14 days. From the 15 th day,all groups except control group were exposed to high altitude hypoxia cabin after 0.5 hour of gavage treatment for 72 hours. After 72 hours,all mice were executed.The content of malondialdehyde(MDA),the activities of total antioxidantcapacity(T-AOC)and super-oxide dismutase(SOD)in lung tissues were detected by colorimetry. Morphological changes of lung issues were observed by HE staining method. Results:Compared with those in control group,the weight of the model group was declined;the indexes of lung,kidney and heart were higher;the MDA content was increased significantly in pulmonary tissue;the pathological score and inflammatoy cells in lung tissues were both significantly increased;the SOD and T-AOC activities of pulmonary tissues were decreased obviously(P〈0.01). Compared with those in model group,the weight was increased in medium and high dose group;the SOD and T-AOC activities were both increased;the indexes of lung,kidney and heart were also decreased;the MDA contents of pulmonary tissue were decreased in medium and high dose group;the pathological score and inflammatoy cells were both decreased in lung tissues(P〈0.05). Conclusion:The astragalus and lily particles could greatly enhance the ability to resistant pulmonary edema in high altitude hypoxia environment,which might be related to the regulation of free radical metabolism.
