摘要:目的观察痛泻要方对肝郁脾虚型溃疡性结肠炎(UC)模型大鼠结肠组织中糖蛋白130(gp130)、细胞因子信号传导抑制蛋白3(SOCS3)基因和蛋白表达的影响。方法将60只大鼠随机分为A、B、C、D组各15只,B、C、D组均采用三硝基苯磺酸/乙醇溶液+束缚应激、饮食失节法行肝郁脾虚型UC造模,A组采用同样方法用等量的生理盐水灌肠。C、D组分别予以痛泻要方(生药5.5 g/kg)、美沙拉秦(0.5 g/kg)灌胃10 m L/kg,A、B组灌服等体积生理盐水。各组造模成功后第1天开始灌胃,1次/d,连续21 d。取出距肛门以上7~8 cm处结肠段,肉眼行结肠黏膜损伤评分,分别采用Real-time荧光定量PCR法及免疫组化、免疫印迹法检测结肠组织中的gp130、SOCS3基因和蛋白。结果 B组结肠黏膜损伤评分为(2.67±0.62)分,高于A组的(0.6±0.63)分(P<0.01);C、D组分别为(1.73±0.70)、(1.27±0.46)分,均高于B组(P均<0.01);C、D组间比较无统计学差异。B组较A组结肠组织中gp130基因和蛋白表达升高、SOCS3基因和蛋白表达降低(P均<0.05),C、D组较B组结肠组织中gp130基因和蛋白表达降低、SOCS3基因和蛋白表达升高(P均<0.05),C、D组结肠组织gp130、SOCS3基因和蛋白比较无统计学差异。结论痛泻要方可下调gp130表达、上调SOCS3表达,从而有效降低肝郁脾虚型UC大鼠结肠黏膜损伤程度。
Objective To observe the effects of Tongxie Yaofang(Tongxie prescription) on the expression of glycoprotein 130(gp130) and suppressor of cytokine signaling 3(SOCS3) gene and protein in colonic tissues of rats with liver depression and spleen deficiency type ulcerative colitis(UC). Methods Sixty rats were randomly divided into groups A,B,C,and D. Rats in the groups B,C,and D were treated with trinitrobenzene sulfonic acid/ethanol solution + restraint stress and improper diet to induce the liver depression and spleen deficiency type UC models. Rats in the group A were treated with the same method by using the same amount of normal saline enema. Rats in the groups C and D were treated with Tongxie Yaofang(crude drug 5. 5 g/kg),mesalazine(0. 5 g/kg) 10 m L/kg by gavage. Rats in the groups A and B were fed with the same volume of normal saline. After the success of modeling in each group,the rats were treated by gavage,once a day for 21 days. We removed the colon segment from 7 to 8 cm above the anus,and determined the colon mucosal injury score by naked eyes. Real-time fluorescent quantitative PCR,immunohistochemistry,and immunoblotting were used to detect gp130,SOCS3 gene and protein in colon tissues. Results The score of colonic mucosal injury in the group B was 2. 67 ± 0. 62,which was higher than that of the group A(0. 6 ± 0. 63)(P〈0. 01); the score of colonic mucosal injury in the group C and group D was 1. 73 ± 0. 70 and 1. 27 ± 0. 46,which was both higher than that of the group B(P〈0. 01). There was no significant difference between groups C and D. Compared with group A,the expression of gp130 gene and protein in the group B was higher and the expression of SOCS3 gene and protein was lower(all P〈0. 05). Compared with group B,the expression of gp130 gene and protein in the groups C and D was lower and the expression of SOCS3 gene and protein was higher than that of group B(all P〈0. 05). There was no significant difference in gp130 and SOCS3 gene and protein between the groups C and D. Conclusion Tongxie Yaofang can inhibit the expression of gp130 and up-regulate the expression of SOCS3,so as to improve the degree of colonic mucosal injury of rats with liver depression and spleen deficiency type UC.
