摘要:目的研究纤维素酶对当归挥发油提取率的影响及化学成分的变化。方法通过单因素考察和正交实验筛选出加酶提取当归挥发油的最佳提取工艺,并利用GC-MS联用技术对其成分进行分离鉴定。结果当归挥发油加酶提取的最佳工艺为加入3%的纤维素酶,在温度为50℃、pH为4.5的缓冲溶液中酶解30min,加水至10倍量,浸泡2.5h,加热回流提取10h。加酶提取法与水蒸气蒸馏法相比,能明显提高当归挥发油得率,可使挥发油得率提升0.31%。GC-MS显示经酶辅助提取后当归挥发油中Z-藁本内酯的含量提高了5.6%。结论加酶提取法工艺简单方便,可显著提高当归挥发油的提取率。
Objective To study the effect of cellulase on the extraction rate and chemical composition of volatile oil from Angelica sinensis.Methods The optimal extraction process for the extraction of volatile oil of Angelica sinensis by enzyme assisted water vapor distillation was selected from single factor examination and orthogonal experiments, then the changes in the extraction yield of volatile oil after addition of enzyme were compared by kinetic model analysis, and the constituents were isolated and identified by GC-MS.Results The optimal extraction procedure for the enzyme assisted extraction of Angelica sinensis volatile oil consisted of the addition of 3% cellulase, enzymatic digestion in a buffer solution with a temperature of 50 ℃, pH 4.5 for 0.5 h, addition of water to 10 fold volume, soaking for 2.5 h, and heating at reflux for 10 h.The best fit to the data by the binomial equation in the kinetic model can achieve high model accuracy, and the fitted results R2 values are all higher than those of the other mathematical models.The enzyme assisted extraction method could obviously improve the yield of volatile oil of Angelica sinensis compared with that of hydrodistillation, and could increase the yield of volatile oil by 0.31%.GC-MS showed that the content of Z-ligustilide in volatile oil of Angelica sinensis increased by 5.6% after enzyme assisted extraction.Conclusion The enzyme assisted extraction method is simple and convenient, which can significantly improve the extraction yield of volatile oil of Angelica sinensis.
