摘要:目的:探讨忍冬藤颗粒对急性痛风性关节炎模型大鼠自体保护作用及炎症因子抑制机制。方法:以乙胺丁醇189mg/kg和腺嘌呤1. 26 g/kg灌胃3周诱导大鼠高尿酸血症模型,在此基础上通过大鼠右踝关节外侧后方穿刺注射尿酸钠溶液0. 2ml/只,连续3天建立大鼠急性痛风性关节炎模型。造模3周后分为23. 8 g/kg、11. 9 g/kg、5. 95 g/kg忍冬藤颗粒组、0. 455 g/kg别嘌呤醇(第3周开始,连续1周)+18. 9 mg/kg秋水仙碱(第4周开始.连续1周)组灌胃给药,连续2周。于干预结束后,用HE染色法观察踝关节软组织炎症表达;采用免疫组化法和PCR法分别观察和检测踝关节软组织HSP70蛋白和基因表达。结果:与空白组比较,模型组大鼠血尿酸水平显著升高,关节液中IL-1β、TNF-α含量显著增加,关节滑膜HSP 70及其mRNA表达量显著增加,有明显的炎症表现;忍冬藤颗粒各组均能不同程度的降低血尿酸值和IL-1β、TNF-α含量,增加HSP 70蛋白及其mRNA表达水平,同时抑制并减轻炎症反应,忍冬藤颗粒23. 8 g/kg、11. 9 g/kg剂量组保护和抑制作用强于5. 95 g/kg剂量组。结论:忍冬藤颗粒降低血尿酸抑制炎症因子、减轻炎症反应的作用可能是通过增强HSP 70表达实现的。
Objective: Objective to investigate the protective effect of honeysuckl-e vine granules on acute gouty arthritis rats and the mechanism of inhibiting i-nflammatory factors. Methods: Rat models of hyperuricemia were induced by e-thambutol(189 mg/kg) and adenine(1. 26 g/kg) for3 weeks,Then the rat mod-el of acute gouty arthritis was established for 3 consecutive days by injecting sodium urate solution(0. 2 ml/rat)through the lateral back of the right sacroilia-c joint of the rat. After 3 weeks of modeling,23. 8 g/(kg·d) high-dose group,11. 9 g/(kg·d)middle-dose group,5. 95 g/(kg·d) low-dose group,0. 455 g/(kg·d) p-urulantol(starting on week 3 for 1 week) + 18. 9 mg/kg colchicine(starting on week 4 and 1 week in a row) for intragastric administration for 2 weeks. Afte-r the intervention,HE staining was used to observe the expression of inflamm-ation in sacroiliac joint soft tissues. Immunohistochemistry and PCR were usedto observe and detect the expression of HSP70 in sacroiliac joint soft tissue. Results: Compared with the blank group,the serum uric acid level in the mode-l group increased,the expression of IL-1β and TNF-α in synovial fluid increa-sed,the expression of HSP70 and mRNA in synovial synovium increased,and there was obvious inflammation. Honeysuckle vine granules can reduce the blo-od uric acid value and the expression of IL-1β and TNF-α in differentdegrees,increase the expression level of HSP70 and mRNA,inhibit and alleviate the i-nflammatory reaction,and the protection and inhibition effect of 23. 8 g/kg and 11. 9 g/kg dose group of lonicera japonicus granules is stronger than that of 5. 95 g/kg dose group. Conclusion: The effect of honeysuckle vine granules on lo-wering serum uric acid and inhibiting inflammatory factors and reducing infla-mmatory reaction may be achieved by enhancing HSP70 expression.
