摘要:目的:分析氧化苦参碱治疗慢性乙型肝炎的作用机制。方法:外周血单个核细胞(PBMCs)分离提取,在体外与氧化苦参碱共孵育48 h,检测Toll样受体7(TLR7)信号通路主要级联分子基因表达水平;TLR7配体刺激经氧化苦参碱预处理的PBMCs 24 h,检测在配体诱导下抗病毒细胞因子分泌水平。结果:氧化苦参碱上调并活化PBMCs细胞中TLR7信号转导通路,在TLR7特异性配体R848刺激下分泌抗病毒细胞因子。结论:氧化苦参碱可上调并活化PBMCs中TLR7信号通路,有利于其发挥其抗CHB作用。
Objective: Investigate the immunomodulatory mechanism of Oxymatrine extracted from Sophora alopecuraides L. to anti-HBV in vitro. Methods: Human peripheral blood mononuclear cells(PBMCs) were isolated from heparinized whole blood from 48 chronic hepatitis B(CHB) patients. The PBMCs from each patient were divided into two groups according to pre-treatment with Oxymatrine or not in vitro. We examined the changes of expression and function of toll-like receptor 7(TLR7) signaling transduction pathway in the PBMCs with different treatment methods and we investigated the TLR7 ligand on antiviral cytokine secretions in vitro. Results: For the TLR7 signaling pathway,Oxymatrine not only augmented the expressions of TLR7 signaling transduction molecules,but also activated the TLR7 signal function.This study clearly demonstrated that TLR7 ligand could stimulate PBMCs that have been pre-treated with Oxymatrine.Furthermore,the quantity of antiviral cytokines secreted by the pre-treated PBMCs was greater than those without pre-treatment. Conclusion: This study suggests Oxymatrine could be a strong immunomodulator,influence TLR7 signaling transduction and improve the immune efficacy of TLR7 ligand against CHB. The data indicates that Oxymatrine possesses an anti-HBV activity and exerts its anti-virus effects via activation of TLR7 signal pathway.
