摘要:目的研究敦煌医方四时常服方对镉染毒大鼠肾氧化应激的影响。方法将60只Wistar大鼠随机分为空白组,模型组,阳性对照组,四时常服方高、中、低剂量组6组,每组10只。空白组腹腔注射0.9%氯化钠注射液,其他5组均腹腔注射0.1%氯化镉溶液1.5 mg/kg,制备镉染毒大鼠模型,每周5次,共5周,染毒的同时分别给予相应药物进行干预,每天1次,连续灌胃5周后处死大鼠,取血和肾组织,检测血清谷草转氨酶(AST)活性及肾组织中超氧化物歧化酶(SOD)活性和丙二醛(MDA)含量,并测定肾脏指数。结果与空白组比较,模型组肾组织中SOD活性明显降低(P<0.01),肾脏指数、肾组织中MDA含量和血清AST活性明显升高(P<0.01);与模型组比较,四时常服方各剂量组肾组织中SOD活性明显升高(P<0.01),肾脏指数、肾组织中MDA含量和血清AST活性均明显降低(P<0.01),其中对SOD,MDA的作用以四时常服方高剂量组最为显著(P<0.05)。结论敦煌医方四时常服方对镉染毒大鼠肾氧化应激具有一定的拮抗作用,可缓解大鼠染镉中毒后肾脏的损伤。
Objective To investigate the effect of Dunhuang Sishi Changfu Fang on renal oxidative stress in rats infected by cadmium. Methods 60 Wistar rats were randomly divided into 6 groups: blank group,model group,positive control group,the high-,medium-and low-dose group of the medical prescription of Sishi Changfu Fang,10 rats in each. The blank group was intraperitoneally injected with 0.9% NaCl solution,and the remaining five groups were intraperitoneally injected with 0.1% cadmium chloride solution( 1.5 mg/kg) for modeling,5 times a week,lasting for 5 weeks. At the same time,the rats exposed to cadmium were given drug intervention for treatment once a day. 5 weeks later,blood and kidney tissues were taken out to detect the contents of aspartate aminotransferase( AST),superoxide dismutase( SOD) and malondialdehyde( MDA). Renal index was determined.Results Compared with blank group,the activity of SOD significantly decreased( P〈0.01),rena index,the content of MDA and the activity of AST increased( P〈0.01) in model group,the differences were statistically significant;compared with model group,the activity of SOD increased( P〈0.01),renal index,the content of MDA,the activity of AST decreased( P〈0.01) in different doses groups of Sishi Changfu Fang,while was most effective on SOD and MDA in the high-dose group( P〈0.05). Conclusion The Sishi Changfu Fang has antagonistic effect on oxidative stress in renal function to alleviate the injury of kidney in rats infected by cadmium.
