摘要:目的:研究附子-干姜药对对慢性溃疡性结肠炎(UC)模型小鼠的保护作用,及对结肠核因子-κB(NF-κB)及炎症因子白细胞介素(IL)-1β、IL-6、IL-10等表达的影响。方法:将60只C57BL/6雄性小鼠随机分为正常组,模型组,固本益肠片组,附子-干姜高、中、低剂量组(6.06、3.03、1.51 g/kg),每组10只,采用2%DSS水溶液制备慢性UC模型,造模30 d,第31天开始灌胃给药,给药体积均为0.2 mL/10 g,连续灌胃10 d,1 d/次。每天记录小鼠体质量、大便性状及便血情况,进行疾病活动指数(DAI)评分。末次给药结束后,取结肠直尺测量结肠长度,肉眼观察肠黏膜炎症损伤程度进行(CMDI)评分。HE染色观察结肠病理组织变化,ELISA检测各组小鼠血清中IL-1β、IL-6、IL-10含量,免疫组化法检测各组小鼠结肠组织NF-κB的表达。结果:与模型组比较,附子-干姜各剂量组小鼠的DAI评分均显著下降(P<0.01),血清中IL-1β含量显著下降(P<0.01)。附子-干姜高、中剂量组CMDI评分显著降低(P<0.01),结肠长度显著增加(P<0.01),结肠黏膜炎症细胞浸润明显减少,腺体结构趋于完整,增生组织明显减少。血清中IL-6含量显著下降(P<0.01),IL-10含量显著升高(P<0.01),结肠组织NF-κB表达含量显著降低(P<0.01)。结论:附子-干姜药对可通过抑制慢性UC模型小鼠结肠组织NF-κB的表达,降低血清中IL-1β、IL-6含量,增加血清IL-10含量减轻慢性UC结肠黏膜炎症。
Objective:To study the protective effect of Aconite-dried ginger on chronic ulcerative colitis(UC)model mice and the effects on the expressions of NF-κB and inflammatory factors IL-1β,IL-6 and IL-10.Methods:A total of 60 male C57BL/6 mice were randomly divided into 6 groups,including normal group,model group,Gubenyizang tablet group,and monksh-ginger high-dose,medium-dose,and low-dose groups(6.06,3.03,1.51 g/kg);with 10 mice in each group.Chronic UC model was prepared by 2%DSS solution for 30 days,and gavage administration was started on the 31st day.The dosage volume was 0.2 mL/10 g,and continuous intragastric administration for 10 d,1 d/time.Body weight,stool characteristics,and blood in stool were recorded every day,and disease activity index(DAI)was scored.After the last administration,the colon was taken for visual observation of the degree of intestinal mucosal inflammation and injury(CMDI)score,and the length was measured.The pathological changes of the colon were observed by hematoxylin-eosin(HE)staining,the expression levels of IL-1β,IL-6,and IL-10 in serum of mice in each group were detected by enzyme-linked immunosorbent assay(ELISA),and the expression levels of NF-κB in colon tissues of mice in each group were detected by immunohistochemistry.Results:Compared with the model group,DAI score and IL-1βcontent in serum were significantly decreased in Aconite-dried ginger three dose groups(P<0.01).CMDI score was significantly decreased(P<0.01),colon length was also improved(P<0.01),inflammatory cell infiltration of colon mucosa was significantly reduced,glandular structure tended to be intact,and hyperplasia tissue was significantly reduced in both high and medium dose groups.Serum IL-6 content was significantly decreased(P<0.01),IL-10 content was significantly increased(P<0.01),and the expression of NF-κB in colon tissue was significantly decreased(P<0.01).Conclusion:Aconite-dried ginger can inhibit the expression of NF-κB in colon tissue of mice with chronic UC,reduce the content of IL-1βand IL-6 in serum,and increase the content of IL-10 in serum to alleviate chronic UC colonic mucosal inflammation.
