摘要:为探究有机肥沼液提升蒙古黄芪品质的分子机理,本研究以一年生蒙古黄芪为材料,利用大田试验,优选沼液处理(biogas slurry,BS),以常规化肥(conventional fertilizer,CF)和清水处理(clear water treatment,CK)为对照,考察不同处理方式对收获期蒙古黄芪生物量、生理特性及根总黄酮(total flavonoids,TAF)、总皂苷(total saponins,TANS)含量的影响,并对3种处理方式下的蒙古黄芪根进行转录组学分析。结果表明,相比CK、CF处理,BS处理可显著增加蒙古黄芪的株高(plant height,PH)及根干重(root dry weight,DWT),提高过氧化物酶(peroxidase,POD)、过氧化氢酶(catalase,CAT)、抗坏血酸过氧化物酶(ascorbic acid peroxidase,APX)活性及游离脯氨酸(free proline,Pro)、可溶性糖(soluble sugar,SS)、可溶性蛋白(soluble protein,SP)的含量,促进其有效成分TAF、TANS含量的积累。转录组分析表明,BS与CK处理组的差异表达基因(differentially expression genes,DEGs)达1755个,与CF处理的DEGs为519个,其调控蒙古黄芪品质的差异基因主要富集在类黄酮、萜类、2-羟基异黄酮还原酶、苯丙烷生物合成等生物学过程,以及淀粉、蔗糖代谢、氧化还原活性、植物激素信号传导途径等有关的分子功能,且每个途径中相关DEGs多数以上调为主。其中APF7、WRKY44、GGT3、CHS、HMGS、UGT73C5可能是沼液提升蒙古黄芪品质的关键调控基因。相关性分析表明,CHS、HMGS、ARF7、WRK44与APX、CAT、TAF、SP、POD、SS、UGT73C5间、UGT73C5与DWT间分别存在极显著正相关关系。研究表明沼液可通过调控蒙古黄芪生物量、生理特性及有效成分生物合成相关基因表达,影响其品质。
This study aims to explore the mechanism of biogas slurry effect on the quality formation of Astragalus membranaceus(Fisch.)Bge.var.mongholicus(Bge.)Hsiao.One year old A.membranaceus seedlings were used as the experimental material.Using a large field experiment,three treatments,namely,the optimized treatment of biogas slurry(BS),conventional fertilizer(CF)and clear water treatment(CK),were set up to investigate the changes in the contents of total flavonoids(TAF)and total saponins(TANS)in roots,biomass and physiological characteristics in roots and leaves.And the transcriptome of A.membranaceus in different treatments was also analyzed.Results showed that compared with CK and CF treatments,BS treatment significantly increased the plant height(PH)and root dry weight(DWT)of A.membranaceus,increased the activities of peroxidase(POD),catalase(CAT)and ascorbic acid peroxidase(APX)and the contents of free proline(Pro),soluble sugar(SS),and soluble protein(SP),as well as promoted the contents accumulation of TAF and TANS.Transcriptome analysis revealed that the number of differentially expression genes(DEGs)between the BS and CK treatment groups amounted to 1755.The DEGs of BS and CF treatments were 519.DEGs were mainly clustered into biological processes such as flavonoids,terpenoids,2-hydroxyisoflavone reductase and phenylpropane biosynthesis,as well as in molecular functions related to starch,sucrose metabolism,redox activity,and phytohormone signaling pathways,etc.And most of the relevant DEGs in each pathway were mainly up-regulated.APF7,WRKY44,GGT3,CHS,HMGS and UGT73C5 genes may be involved in the process of improving quality of A.membranaceus by biogas slurry.Correlation analysis showed highly significant positive correlations between CHS,HMGS,ARF7,WRKY44 and APX,CAT,TAF,SP,POD,SS,UGT73C5.Similarly,there were significant positive correlations between UGT73C5 and DWT.This study indicated that biogas slurry can regulate the biomass,physiology and active ingredient biosynthesis of A.membranaceus to promote the formation of its quality.
