摘要:目的探讨小檗碱(berberine,BBR)对大鼠痤疮模型皮损及Toll样受体(TLR)4/核因子κB(nuclear factor kappa-B,NF-κB)信号通路的影响。方法将48只SD大鼠随机分为空白组、模型组、甲硝唑凝胶组及小檗碱低、中、高剂量组,除空白组外其余各组均建立大鼠耳廓痤疮模型,造模21 d。造模结束后,空白组不予任何处理,模型组给予空白凝胶基质,甲硝唑凝胶组外用甲硝唑凝胶0.010 g/(cm^(2)·d),小檗碱低、中、高剂量组分别外用小檗碱0.005、0.010、0.020 g/(cm^(2)·d),治疗28 d。观察治疗前后皮损变化;HE染色观察大鼠耳廓皮损组织病理变化;RT-PCR法检测大鼠耳廓皮损组织中TLR4、NF-κB、P38MAPK mRNA表达水平;Western blot法检测大鼠耳廓皮损组织中TLR4、NF-κB、P38MAPK、ERK蛋白表达水平。结果治疗后,小檗碱组观察到表皮角化程度明显改善,棘层变薄,真皮浅层炎细胞浸润明显改善等;对比空白组,模型组皮损组织中TLR4、NF-κB、P38MAPK mRNA及蛋白表达水平升高,差异有统计学意义(P<0.05);ERK蛋白表达水平升高,差异有统计学意义(P<0.05)。对比模型组,甲硝唑凝胶组及小檗碱高、中剂量组中的TLR4、NF-κB、P38MAPK mRNA表达水平均有不同程度的降低,差异有统计学意义(P<0.05);小檗碱低剂量组中的TLR4、NF-κB mRNA虽有一定程度降低,但差异无统计学意义(P>0.05);甲硝唑凝胶组及小檗碱高、中剂量组中的TLR4、NF-κB、P38MAPK、ERK蛋白表达水平均降低,差异有统计学意义(P<0.05)。结论外用小檗碱可显著改善大鼠耳廓痤疮模型的皮损,其作用可能与降低局部组织TLR4、NF-κB、P38MAPK、ERK的表达相关。
Objective To investigate the effects of berberine(BBR)on skin lesions and Toll-like receptor(TLR)4/nuclear factor kappa-B(NF-κB)signaling pathway in rat acne model.Methods Forty-eight SD rats were randomly divided into blank group,model group,metronidazole gel group,berberine low dose group,medium dose group and high dose group.The rat auricular acne model was established in all groups except the blank group for 21 days.After modeling,the blank group received no treatment,the model group received blank gel matrix,the metronidazole gel group was treated with metronidazole gel 0.010 g/(cm^(2)·d),and the berberine groups received topical berberine at 0.005,0.010 and 0.020 g/(cm^(2)·d)respectively for 28 days.Skin lesion changes were observed before and after treatment.Pathological changes in rat auricles were examined by HE staining.RT-PCR was used to detect mRNA expression levels of TLR4,NF-κB and P38MAPK in auricular lesions.Western blot was performed to measure protein expression levels of TLR4,NF-κB,P38MAPK and ERK.Results After treatment,the berberine groups showed significant improvement in epidermal keratinization,thinning of the stratum spinosum,and reduced inflammatory cell infiltration in the superficial dermis.Compared with the blank group,the model group exhibited increased mRNA and protein expression levels of TLR4,NF-κB and P38MAPK(P<0.05),as well as elevated ERK protein expression(P<0.05).Compared with the model group,the metronidazole gel group and berberine medium-and high-dose groups showed decreased mRNA expression levels of TLR4,NF-κB and P38MAPK(P<0.05).The berberine low-dose group showed some reduction in TLR4 and NF-κB mRNA levels,but this was not statistically significant(P>0.05).Berberine medium-and high-dose groups exhibited decreased protein expression levels of TLR4,NF-κB,P38MAPK and ERK(P<0.05).Conclusion Topical berberine can significantly improve skin lesions in rat acne model,possibly by downregulating the expression of TLR4,NF-κB,P38MAPK and ERK in local tissues.
