摘要:目的:观察平痫冲剂对戊四唑点燃幼龄癫痫模型大鼠血脑屏障病理结构及相关蛋白Cx43、ICAM-1表达的影响。方法:抽选60只空白大鼠为空白组,癫痫模型制备成功后将180只幼龄大鼠随机分为模型组、丙戊酸钠组(0.02 g/mL)及平痫冲剂组(0.19 g/mL),每组各60只,灌胃21天,每天1次,空白组予等量生理盐水灌胃。在灌胃24 h、48 h、72 h、7 d、14 d、21 d取大鼠脑组织,进行血脑屏障超微结构观察、血脑屏障通透性测定及对Cx43、ICAM-1蛋白表达的观察。结果:平痫冲剂可恢复幼龄癫痫模型大鼠的血脑屏障超微结构、降低血脑屏障的通透性、下调血脑屏障相关蛋白Cx43与ICAM-1的表达。结论:平痫冲剂可能通过对血脑屏障结构及通透性的调节,进而发挥抗痫作用。
Objective:To observe the effect of Pingxian granule on pathological structure of blood brain barrier and expressions of related proteins Cx43 and ICAM-1 in young epileptic modelled rat kindled with pentylenetetrazole.Methods:60 blank rats were selected as blank group.After successful preparation of epileptic model,180 young rats were randomly divided into model group,sodium valproate group(0.02 g/mL)and Pingxian granule group(0.19 g/mL);60 rats in each group were given intragastric administration for 21 days once a day.The blank group was given the same amount of normal saline.Brain tissues of rats were taken at 24 h,48 h,72 h,7 d,14 d and 21 d after gavage.The ultrastructure of blood-brain barrier was observed.The permeability of blood-brain barrier was determined.The expressions of Cx43 and ICAM-1 proteins were observed.Results:Pingxian granule could restore the ultrastructure of blood-brain barrier in young epileptic modelled rats,reduce the permeability of blood-brain barrier,and down-regulate the expressions of related proteins Cx43 and ICAM-1 of blood-brain barrier.Conclusion:Pingxian granule may play an anti-epileptic role by regulating the structure and permeability of blood-brain barrier.
